Syncytin、MMP-2、MMP-9的表达与胎儿生长受限的关系
Effects of abnormal Syncytin expression on trophoblast invasiveness and fetal growth restriction
摘要目的 探讨Syncytin表达异常对滋养细胞侵袭能力的影响.方法 选取我院2014年4月至12月的58例胎儿宫内生长受限患者作为试验组,选取58例正常妊娠妇女及胎盘植入患者作为对照组,采用荧光定量PCR和Western Blotting方法检测不同胎盘组织中Syncytin、MMP-2、MMP-9的表达;20只SD受孕大鼠随机分为处理组(静脉注射Syncytin抗体,n=10)和对照组组(n=10,不注射Syncytin),妊娠足月时终止妊娠,比较两组胎鼠重量,采用HE染色,比较两组母鼠胎盘的显微结构及胎盘侵袭子宫的程度,荧光定量PCR、Western blotting检测胎盘组织Syncytin、MMP-2、MMP-9 mRNA和蛋白表达.结果 试验组Syncytin、MMP-2、MMP-9 mRNA和蛋白水平表达水平明显少于对照组(P<0.05),且Syncytin的表达与MMP-2、MMP-9呈正相关(P<0.05);处理组孕足月胎鼠的体重明显小于对照组(P<0.05);处理组Syncytin、MMP-2、MMP-9 mRNA和蛋白水平表达明显少于对照组(P<0.05).结论 胎盘组织Syncytin表达下调与胎儿生长受限发病密切相关,Syncytin基因在胎儿生长受限疾病的发生、发展过程中起重要作用.

Methods From April 2014 to December,58 cases of intrauterine growth retardation patients as experimental group,58 cases of normal pregnant women and placenta implantation patients as control group,used real-time fluorescence quantitative PCR(real-time FQ-PCR)detection and Western-blot method to detect Syncytin,MMP-2,MMP-9 of the placental tissue expression and correlation of Syncytin, MMP-2,MMP-9. 10 SD pregnant rats were given intravenous injection of Syncytin antibody pretreatment, and 10 were not only pre Syncytin antibody Treatments,which were compared to full-term pregnancy termination of pregnancy,weighing treatment group and untreated group of fetal weight by electronic balance,used HE staining,optical microscope group 2 SD rat placental microstructure and invasion degree of uterine placenta,The expression of Syncytin,MMP-2 and MMP-9 in placental tissues of different SD rats were detected by real-time fluorescent quantitative PCR (real-time),Western-blot and other methods respectively. Results The expression level of MMP-2,Syncytin,MMP-2,MMP-9,Syncytin and protein level,MMP-9 mRNA of the experimental group was significantly less than the control group(P<0.05);the expression of Syncytin and MMP-2,the expression of MMP-9 was positively correlated(P<0.05);treatment group of full-term fetal weight was significantly smaller than the untreated (P<0.05);Syncytin,MMP-2, MMP-2,MMP-9,MMP-9 and Syncytin protein level,the expression of mRNA The level of treatment group was significantly less than the control group(P<0.05). Conclusion The down-regulation of syncytin expression in placental tissues is closely related to the onset of fetal growth restriction. Syncytin gene plays an important role in the occurrence and development of fetal growth restriction disease.
