地塞米松对哮喘核苷酸结合寡聚域样受体1和2 mRNA 表达的影响

摘要:

目的:探讨核苷酸结合寡聚域(NOD)样受体在哮喘发病机制中的作用。方法采用哮喘大鼠模型,24只大鼠采用数字表法随机分为哮喘组、对照组和地塞米松组,每组8只,采用实时荧光定量 PCR 法检测肺组织 NOD1和2 mRNA 的表达。结果哮喘组、对照组、地塞米松组 NOD1表达量分别为(0.62±0.34)RQ 值、(1.00±0.00)RQ 值、(0.65±0.33)RQ 值,哮喘组显著低于对照组(F =4.75,P <0.05),地塞米松组表达量与哮喘组差异无统计学意义(P >0.05)。哮喘组、对照组、地塞米松组 NOD2 mRNA 表达量为(0.92±0.32)RQ 值、(1.00±0.00)RQ 值、(1.50±0.56)RQ 值,哮喘组 NOD2表达量与对照组差异无统计学意义(P >0.05),地塞米松组 NOD2表达量显著高于哮喘组(F =5.64,P <0.01)。NOD1和2 mRNA 的表达水平无显著相关性(n =24,r =0.15,P >0.05)。结论不同亚型的 NOD 样受体在哮喘大鼠肺组织中的表达不同,对地塞米松的反应也不相同。

Objective To investigate the potential roles of nucleotide -binding oligomerization domain (NOD)-like receptor in the pathogenesis of asthma.Methods Through rat asthma model,24 rats were randomly divided into three groups on average,named asthma group,control group and dexamethasone group.Expression levels of NOD1 and NOD2 mRNA were detected by Real -time PCR in lung tissues.Results The expression levels of NOD1 mRNA in the asthma group,control group and dexamethasone group were (0.62 ±0.34)RQ value,(1 .00 ± 0.00)RQ value,(0.65 ±0.33 )RQ value respectively.The levels of NOD1 mRNA in the asthma group was significantly lower than that in the control group(F =4.75,P <0.05 ),while there was no statistically significant difference of NOD1 mRNA level between the dexamethasone group and asthma group(P >0.05).Moreover,expression levels of NOD2 mRNA in the asthma group,control group and dexamethasone group were (0.92 ±0.32)RQ value, (1 .00 ±0.00)RQ value,(1 .50 ±0.56)RQ value,respectively.There was no statistically significant difference of NOD2 mRNA level between the asthma group and control group (P >0.05 ),but level of NOD2 mRNA in the&nbsp;dexamethasone group was significantly higher than that in the asthma group(F =5.64,P <0.01 ).And there was no significant correlation between level of NOD1 and NOD2 mRNA(r =0.1 5,P >0.05).Conclusion Expression of NOD -like receptor subtype was not at the same level,and their reaction to dexamethasone were different either.
