不同浓度丁酸钠体外诱导树突状细胞分化的细胞表型研究
The cell phenotypes of different sodium butyrate-induced human monocyte-derived dendritic cells in vitro
目的 探讨不同浓度丁酸钠诱导对体外培养树突状细胞(DC)细胞表型的变化规律.方法 通过粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和白细胞介素-4(IL-4)体外诱导的人外周血单个核细胞来源的DC,按不同丁酸钠浓度和不同诱导时间分组培养,流式细胞仪检测各组DC的细胞表型.结果 在丁酸钠诱导下,各组DC表面的成熟标志均显著下调,丁酸钠浓度越高下调越明显,随着培养时间的延长,各组DC表面的成熟标志均显著下调;但是细胞的凋亡率也同时上升.结论 丁酸钠在体外可以显著抑制DC的成熟过程,以丁酸钠浓度为0.75mmol/L和培养48h为最佳的诱导浓度和时间.
更多Objective To investigate the cell phenotypes of different sodium butyrate--induced human monocyte-derived dendritic cells (DCs) in vitro.Methods The human monocyte-derived DCs were induced in the presence of cytokine recombined human GM-CSF and interleukin (IL)-4.After culture with different concentrations of sodium butyrate for different time,the cell phenotypes of DCs were detected by flow cytometry.Results The sodium butyrate could down-regulate the major histocompatibility complex (MHC) class II and costimulatory molecules of DCs in a concentration- and time-dependent manner.At the same time the apoptosis rate was increased.Conclusion The sodium butyrate could significantly inhibit the maturation of DCs in vitro.The optimal concentration of sodium butyrate and time for induction was 0.75 mmol/L and 48 h.
