缺氧预处理对神经元、神经干细胞Notch1蛋白表达的影响
Effects of hypoxic preconditioning on expression of Notch1 signal in neurons, and neural stem cells
目的 观察缺氧预处理对神经元、神经干细胞Notch1蛋白表达的影响.方法 将神经元、神经干细胞分成对照组(cell组),缺氧预处理组(a组)、缺血再灌注损伤组(b组)、缺氧预处理+缺血再灌注损伤组(a+b组),Western blot检测各组Notch1表达.结果 神经元、神经干细胞的a组(0.524±0.050、0.104 ±0.030)、b组(0.570±0.100、0.122±0.050)和a+b组(1.294±0.160、0.550±0.150) Notch1蛋白表达均高于cell组(0.394±0.060、0.031 ±0.080),差异有统计学意义(均P <0.05);神经元、神经干细胞的a+b组(1.294±0.160、0.550±0.150) Notch1蛋白表达均高于各自的b组(0.570 ±0.100、0.122±0.050,P<0.05).结论 缺血再灌注损伤和缺氧预处理后,两种细胞Notch1蛋白表达均上调,缺氧预处理使两种细胞缺血再灌注损伤后Notch1蛋白表达上调更明显.
更多Objective To observe the effects of hypoxic preconditioning hypoxic preconditioning on the expression of Notch1 signal in neurons,and neural stem cells.Methods Primary cultured neurons and neural stem cells were divided into four groups:normal cells group (cell group),hypoxic preconditioning group (a group),ischemia-reperfusion injury group (b group),and hypoxic preconditioning + ischemia-reperfusion injury group (a + b group).By Western blotting was used to detect the expression of Notch1 of two types of nerve cells in four groups.Results Notch1 protein expression in neurons and neural stem cells of a group (0.524 ±0.050,0.104 ±0.030),b group (0.570 ±0.100,0.122 ±0.050) group and a + b group (1.294 ± 0.160,0.550 ± 0.150) was statistically significantly higher than that in cell group (0.394 ± 0.060,0.031 ± 0.080,P < 0.05).The Notch1 protein expression in two types of cells in a + b group (neurons 1.294±0.160,neural stem cells 0.550 ±0.150) was significantly higher than that in the respective cell group (0.570 ±0.100,0.122 ±0.050,P<0.05).Conclusion In neurons and neural stem cells after ischemia-reperfusion injury or hypoxic preconditioning,the expression of Notchl signals is upregulated.Mter hypoxic preconditioning plus ischemia-reperfusion injury,the expression of Notch1 signals in the two types of cells is up-regulated more obviously.
