鞣酸对大鼠心室肌场电位时限及兴奋传导速度影响的研究

Effects of tannic acid on the field action potential duration and excitation conduction velocity in rat ventricular myocytes

摘要:

目的 应用微电极阵(MEA)技术研究鞣酸在生理及病理状态下对大鼠心室肌场电位时限(fAPD)及兴奋传导速度(ECV)的影响.方法 大鼠100只随机分成6组,对照组(n=10)、鞣酸组(10、20、40 μmol/L,每种浓度n=10)、海葵毒素组(ATXⅡ,n=10)、ATXⅡ+鞣酸组(10、20、40 μmol/L,每种浓度n=10)、心力衰竭(心衰)组(n=10)、心衰+鞣酸组(40 μmol/L,n=10).开胸取出心脏后进行langendroff灌流,用柔性电极贴附左心室,分别记录各组场电位及兴奋传导的变化.结果 MEA可敏感记录到各组场电位及兴奋传导的变化.鞣酸10 μmol/L组、20 μmol/L组、40 μmol/L组心室肌fAPD分别缩短至(345.70±10.33) ms、(329.30±12.18) ms、(317.20±11.31) ms,与对照组相比差异有统计学意义(P<0.05);ECV分别加快至(0.63±0.04) m/s、(0.66±0.03) m/s、(0.70±0.05) m/s,与对照组相比差异有统计学意义(P<0.05);ATXⅡ组fAPD明显延长(P<0.05),ECV则无明显变化(P>0.05).与ATXⅡ组相比,ATXⅡ+鞣酸10 μmol/L组、20 μmol/L组、40 μmol/L组心室肌fAPD分别缩短至(372.20±13.18) ms、(337.50±1Z 28) ms、(317.40±11.49) rs,差异有统计学意义(P<0.05),ECV分别加快至(0.62±0.03) m/s、(0.66±0.04) m/s、(0.72±0.03) m/s,差异有统计学意义(P<0.05),其fAPD缩短率明显高于鞣酸组.与对照组相比,心衰组fAPD延长、ECV减慢,差异有统计意义(P<0.05).与心衰组相比,心衰+鞣酸组fAPD缩短至(441.70±13.81) ms、ECV加快至(0.55±0.06) m/s,差异有统计学意义(P<0.05).结论 应用MEA技术可真实、准确地反映心室肌电生理特性.鞣酸在生理及病理状态下均可缩短大鼠心室肌fAPD及加快ECV,显示出其可能具有较好的抗心律失常及治疗心衰等作用.

Objective To explore the effects of tannic acid on the field action potential duration (fAPD)and excitation conduction velocity(ECV) in rat ventricular myocytes under physiological and pathological conditions by microelectrode arrays(MEA) technology.Methods One hundred rats of either sex were randomly divided into 6 groups:control group(n =10),tannic acid group (10,20,40 μmol/L,each concentration n =10),ATX Ⅱ group (n=10),ATX Ⅱ +tannic acid group (10,20,40 μmol/L,each concentration n=10),heart failure group(n =10),and heart failure+tannic acid group (n =10).Flexible electrodes were attached to these hearts quickly removed and perfused with modified Tyrode's solution.Changes of fAPD and ECV in each group were recorded.Results The changs of fAPD and ECV in each group were sensitively recorded by MEA.In tannic acid 10 μmol/L group,20 μmol/L group,and 40 μmol/L group,fAPD were shortened to (345.70 ±10.33) ms,(329.30±12.18) ms,and (317.20±11.31) ms(P<0.05);ECV were accelerated to (0.63±0.04)m/s,(0.66±0.03) m/s,and(0.70±0.05) m/s(P<0.05) compared with the control group.In ATXⅡ group,fAPD obviously prolonged (P<0.05) and ECV had no change(P<0.05)compared with the control group.In ATXⅡ+tannic acid 10 μmol/L group,20 μmol/L group,and 40 μmol/L group,fAPD were respectively shortened to (372.20±13.18) ms,(337.50±12.28) ms,and(317.40±11.49) ms(P<0.05);ECV were respectively accelerated to (0.62±0.03) m/s,(0.66±0.04) m/s,and (0.72 ± 0.03) m/s (P < 0.05);and the shortening rate was significantly higher than that of tannin group(P<0.05).In heart failure group,fAPD prolonged and ECV slowed compared with the control group (P<0.05).In heart failure+tannic acid group,fAPD was shortened to (441.70±13.81) ms and ECV was accelerated to (0.55±0.06) m/s compared with the heart failure group (P<0.05).Conclusion MEA is a sensitive marker for the measurement of the ventricular myocytes electrophysiological propeaies.Tannic acid can concentration-dependently shortened fAPD and accelerated ECV in rat ventricular myocytes under physiological and pathological conditions,and may play a role in antiarrhythmias and heart failure.
