卡托普利、氟伐他汀对博莱霉素致肺纤维化大鼠的实验研究
Experimental study on captopril and fluvastatin in bleomycin induced pulmonary fibrosis in rats
目的 观察卡托普利、氟伐他汀对博莱霉素所致的大鼠肺纤维化的治疗作用,探讨治疗肺纤维化的新途径.方法 将50只Wistar大鼠一次性气管内注射博莱霉素造成肺纤维化模型,随机分为4组.博莱霉素模型组(n=15):大鼠气管内一次性注入博莱霉素5mg/kg,制成肺间质纤维化模型;卡托普利治疗组(n=15);氟伐他汀治疗组(n=15);对照组(n=5).各组动物均于气管注射后第7、14、28天取肺组织,行病理HE、Masson染色,转化生长因子β1检测以及第28天测定肺系数.结果 卡托普利治疗组及氟伐他汀治疗组各期肺组织转化生长因子β1含量较博莱霉素模型组降低(P<0.05或P<0.01),肺系数减少(P<0.05),肺组织病理学显示肺泡炎及肺纤维化程度均明显减轻.结论 卡托普利及氟伐他汀对肺间质纤维化有良好的防治作用.
更多Objective To investigate the therapeutic effect of captopril and fluvastatin on bleomycin induced pulmonary fibrosis in rats and explore a new way of curing pulmonary fibrosis.Methods 50 Wistar rats were replicated into pulmonary fibrosis models by intratracheal injection in a single of bleomycin A5 and were divided into model group(n =15),captopril-treated group(n =15),fluvastatintreated group(n =15),and control group(n =5)randomly.Two groups were administered intratracheal injection of fluvastatin and captopril.Model group and control group rats were administered saline.Five rats of every group were put to death and made section of lungs with HE or Masson staining and immunohistochemistry after 7,14,28 days.On the 28th day,lung coefficients of each group rats were compared.Results Transforming growth factor-β1 contents in the captopril-treated and fluvastatin-treated group were obviously lower than those in model group at all phases(P <0.05 or P <0.01),and so was the lung coefficients.The degree of alveolitis and fibrosis of lung studied by histopathological in the captopril-treated group and fluvastatin-treated group was lower than that in model group.Conclusions Captopril and fluvastatin has satisfactory curative effect on pulmonary fibrosis.
