COX-2、ICAM-1、IFN-γ在慢性前列腺炎大鼠血清中的表达及意义
Expression and significance of COX-2, ICAM-1 and IFN-γin serum of rats with chronic pros-tatitis
摘要目的 探究环氧化酶-2(COX-2)、细胞间粘附分子-1(ICAM-1)、γ-干扰素(IFN-γ)在慢性前列腺炎大鼠血清中的表达及意义.方法 选取45例由佳木斯大学动物研究中心提供的Wistar雄性大鼠,随机分为对照组和模型组,其中模型组大鼠构建慢性前列腺炎模型.使用Western blot法检测COX-2蛋白、ICAM-1蛋白、IFN-γ蛋白表达水平,使用RT-PCR检测COX-2 mRNA、ICAM-1 mRNA、IFN-γmRNA的表达水平.结果 对照组和模型组COX-2、ICAM-1、IFN-γ蛋白及mRNA均表达一致.模型组COX-2蛋白、COX-2 mRNA表达水平均显著高于对照组(P<0.05);模型组ICAM-1蛋白、ICAM-1 mRNA表达水平均显著高于对照组(P<0.05);模型组IFN-γ蛋白、IFN-γmRNA表达水平均显著高于对照组(P<0.05);三项指标单独检测时,IFN-γ的敏感度、特异度和准确度均高于其他两项,三项联合检测时,敏感度、特异度、准确性显著高于单独检测,差异有统计学意义(P<0.05).前列腺素的表达随着COX-2表达的增多而增多,慢性前列腺炎的不同期型对其分泌也有影响.结论 COX-2、ICAM-1、IFN-γ表达水平变化与慢性前列腺炎有密切关联,检测COX-2、ICAM-1、IFN-γ的表达水平变化对慢性前列腺炎的诊断、治疗及预后都有着极其重要的意义.

abstractsObjective To investigate the expression and significance of Cyclooxygenase-2 (COX-2), Intercellular adhesion molecular-1 (ICAM-1), Interferon-γ(IFN-γ) in serum of chronic prostatitis rats. Methods Forty-five male Wistar rats were randomly divided into the control group and the model group, which were provided by the animal research center of Jiamusi university. The model group was constructed with the model of chronic prostatitis. The expression level of COX-2 pro-tein, ICAM-1 protein and IFN-γ protein was detected by Western blot, and the expression level of COX-2 mRNA, ICAM-1 mRNA and IFN-gamma mRNA was detected by RT-PCR. Results The ex-pression of COX-2, ICAM-1, IFN-γprotein and mRNA in the control group and the model group were consistent. The expression level of COX-2 protein and COX-2 mRNA in the model group was signifi-cantly higher than that in the control group(P<0. 05), and the expression level of ICAM-1 protein and ICAM-1 mRNA in the model group was significantly higher than that of the control group( P<0. 05), and the expression level of IFN-γprotein and IFN-γmRNA in the model group was significantly higher than that of the control group(P<0. 05). The sensitivity of the three indexes was detected. Sensitivity, specificity and accuracy were all higher than those of the other two items. The sensitivity, specificity and accuracy of the three joint tests were significantly higher than those of the single test, with a statistically significant difference(P<0. 05). The expression of prostaglandin increased with the increase of COX-2 expression, and the different stages of chronic prostatitis also had an effect on its se-cretion. Conclusions The changes of expression level of COX-2, ICAM-1 and IFN-γ are closely re-lated to chronic prostatitis. It is very important for the diagnosis, treatment and prognosis of chronic prostatitis to monitor the changes of expression level of COX-2, ICAM-1 and IFN-γ.
