目的 体外培养健康人及隐睾和尿道下裂患儿尿源性细胞并对其生物学特性进行鉴定.方法 分别收集健康人、隐睾和尿道下裂患儿的清洁尿液,离心后收集沉渣,接种培养,连续传代.对培养得到的细胞进行细胞免疫荧光染色、核型分析、短串连重复序列分析等实验鉴定其生物学特性.结果 倒置相差显微镜下观察细胞的生长,细胞在体外培养条件下增殖旺盛,根据其传代时的细胞数绘制的生长曲线显示:隐睾和尿道下裂患儿的尿液来源的细胞与健康人尿液来源的细胞具有相似的体外增殖能力；细胞免疫荧光染色显示:尿源性细胞表达尿道上皮细胞表面标志物；这些细胞均保持正常的核型；短串连重复序列实验表明:这些细胞分别具有自身独特的基因座,其DNA分型在ATCC、JCRB、DSMZ细胞库中均未找到与其细胞分型100％相匹配的细胞.结论 无论是健康人还是疾病患儿的尿液中均能培养出大量细胞,这些细胞以上皮细胞为主；这些细胞核型正常,携带有相应个体独一无二的遗传信息,可作为简便无创的细胞来源,为研究个体特异的、疾病特异的机制研究和临床诊断提供来源.

Objective To culture and identify the cells from urine of healthy people and children with cryptorchidism. Methods Cleaning urine were harvested and centrifuged for collection of pellet, then followed by seeding and propagation. Cell counting, immunofluorescene staining, and karyotyping, as well as short tandem repeats analysis were conducted to identify the characteristics of cells. Results Urine-derived cells growth curve showed that cells arising from healthy people and children with cryptorchidism and grew well and presented a similar propagation capacity. ICC staining suggested these cells expressed urothelial surface markers. Furthermore, these cells displayed normal karyotype and cells had their specific short tandem repeats. Conclusions The urine is an ideal non-invasive cell source that can be utilized for not only individual-specific and disease-specific mechanism researches, but also for clinical diagnosis of differential individuals.
