肿瘤细胞裂解物致敏的树突状细胞对卵巢上皮癌疗效的体外研究
Tumor cells lysates pulsed dendritic cells for immunotherapy of ovarian cancer in rats: in-vitro study
目的 探讨肿瘤细胞裂解物致敏树突状细胞(DC)疫苗对卵巢癌治疗的可行性.方法 采取随机对照研究方法 ,将大鼠分成实验组与对照组,每组20只,每组随机抽取10只大鼠进行DC体外培养,其余大鼠随后取脾脏淋巴细胞进行杀伤率试验,实验组于培养的第3天加入大鼠卵巢癌细胞系NuTu-19的冻融抗原,培养14 d后将两组DC分别与各组剩余大鼠脾脏分离的T淋巴细胞共同培养,观察形态学.结果 实验组DC在体外试验中可以有效地刺激T淋巴细胞的增殖,诱导生成杀伤性T淋巴细胞(CTL),在效靶比(SC:RC)为1:3、1:10、1:30和1:100时:淋巴细胞刺激指数(SI)实验组均高于对照组(P<0.01);在相同效靶比(SC:RC=3:1,10:1,30:1)时:对肿瘤细胞的杀伤率实验组分别为(40.9±1.2)%、(69.8±1.8)%及(89.0±0.4)%,对照组分别为(30.9±1.9)%、(34.4±2.1)%及(51.0±1.5)%,两者比较差异具有统计学意义(P<0.01).结论 肿瘤细胞裂解物致敏后的DC疫苗能够激活T淋巴细胞,对肿瘤细胞产生体外免疫杀伤作用.
更多Objective To study the feasibility of ovarian tumor cells lysates pulsed dendritic cells (DEs) in the immunotherapy of tumor prevention via cytotoxic test in-vitro. Methods Forty rats were randomly divided into 2 gropups: experimental and control groups. Ten rats from each group were killed and DCs were isolated and cultured. Frozen-thawed antigen of in the study group, rat's Ovarian cancer cells of the line NuTu-19 were cultured, freeze-meh antigens were added at day 3. Two groups co-cultured with T lymphocytes from the other rat's spleen on the 14th day. Then observed the shape of DCs, contrasted the proliferation and killing rate. Results In the study group, DCs could significantly stimulate T lymphocytes multiplication and produce tumor specific cytotoxic T lymphocytes (CTL). When the ratios of stimulating cells (SC) to reaction cells (RC) were 1: 3,1: 10,1:30 and 1: 100,the stimulating index (SI) in the study group were much higher than those in control group (P<0.01). When the ratios of SC is the same with RC (SC: RC=3: 1,10: 1,30: 1) :the killing and wounding rate to tumor cells of study group [(40.9±1.2)%, (69.8±1.8)%, (89.0±0.4%)] is significantly higher than those of contol group [(30.9±1.9)%, (34.4±2.1)%, (51.0±1.5)%] (P<0.01). Conclusion DC could activate T lymphocytes proliferation and produce CTL to kill and wound tumor cells in-vitro.
