摘要二甲双胍是目前临床应用最为广泛的2型糖尿病治疗药物;其主要通过抑制肝糖输出和改善胰岛素敏感性而发挥降糖作用.此外,越来越多的研究表明,二甲双胍除降糖作用之外,还有抗炎、抗癌和抗衰老作用等.目前有关二甲双胍抗炎、抗癌和抗衰老作用的机制尚未明确,但研究表明,其主要涉及AMPK途径的激活.进一步加强对二甲双胍抗炎、抗癌和抗衰老作用及其机制的研究,将有助于开拓二甲双胍临床应用的新领域.

abstractsCurrently,metformin is the most-widely clinicall used drug for type 2 diabetes;it improves hyperglycemia primarily by suppressing hepatic glucose production and increasing insulin sensitivity.In addition,accumulating evidences suggest that,beside glucose-lowering effect,metformin also possesses the properties of anti-inflammatory,anti-cancer,and anti-aging effects.Although the underlying mechanisms of anti-inflammation,anti-cancer,and anti-aging currently remain to be elucidated,studies indicate that it primarily involves the activation of AMPK pathway.Further study on the effects and mechanisms will be beneficial to exploiting the new field of the clinical use of metformin.
