新加归肾丸对多囊卵巢综合征模型大鼠卵泡发育障碍的影响
Effects of Xinjia Guishen pills on impaired follicular development in rat models with polycystic ovarian syndrome
摘要目的 探讨新加归肾丸对多囊卵巢综合征(PCOS)模型大鼠卵泡发育障碍的影响.方法 30只雌性SD大鼠利用随机数字表进行完全随机化分为对照组、模型组和新加归肾丸组,每组10只.模型组和新加归肾丸组均皮下注射人绒毛膜促性腺激素和中性胰岛素,制备大鼠PCOS模型,对照组皮下注射等量生理盐水.新加归肾丸组灌胃新加肾归丸0.23 g·kg-1·d-1,连续灌胃给药14d,模型组和对照组灌胃等量生理盐水.治疗后,检测各组大鼠血清性激素、抗苗勒氏管激素(AMH)、胰岛素样生长因子(IGF-I)、肿瘤坏死因子(TNF-α)和血管内皮生长因子(VEGF)水平,HE染色观察卵巢组织形态.结果 与对照组相比,模型组大鼠睾酮(T)、雌激素(E2)、AMH、IGF-I、TNF-α和VEGF水平显著升高(P<0.05),成熟卵泡数量显著减少(P<0.05);与模型组相比,新加归肾丸组大鼠T、E2、AMH、IGF-I、TNF-α和VEGF水平显著下降(P<0.05),成熟卵泡数量显著增加(P<0.05).结论 新加归肾丸可以有效调节PCOS大鼠体内的性激素水平,抑制胰岛素抵抗和VEGF的表达,促进卵泡的发育成熟.

abstractsObjective To investigate the effects of Xinjia Guishen pills on the impaired follicular development in the rat models with polycystic ovarian syndrome (PCOS).Methods Thirty SD rats were randomly divided into the control group,model group,and Xinjia Guishen pills group (v=10 each).The model group and Xinjia Guishen pills group were injected subcutaneously with human chorionic gonadotropin (HCG) and neutral insulin to prepare rat PCOS model,and the control group was injected subcutaneously with the same amount of normal saline.The Xinjia Guishen pills group was intragastrically given 0.23 g · kg-1 · d-1 Xinjia Guishen pills for continuous administration of 14 days.The model group and control group were intragastrically given the same amount of normal saline.After the treatment,the serum levels of sex hormones,anti-Mullerian hormone (AMH),insulin-like growth factor (IGF-I),tumor necrosis factor (TNF-α) and vascular endothelial growth factor (VEGF) were measured.HE staining was performed to determine the morphology of ovarian tissue.Results Compared with the control group,the levels of testosterone (T),estrogen (E2),AMH,IGF-I,TNF-α and VEGF in the model group significantly increased (P<0.05),whereas the number of mature follicles significantly decreased (P<0.05).Compared with model group,the levels of T,E2,AMH,IGF-I,TNF-α and VEGF in Xinjia Guishen pills group significantly decreased (P<0.05),whereas the number of mature follicles significantly increased (P<0.05).Conclusion:Xinjia Guishen pills may effectively regulate the levels of sex hormones in PCOS rats,inhibit the expression of insulin resistance and VEGF,and promote the development and maturation of follicles.
