亚砷酸钠对PC12细胞色素上皮衍生因子及凋亡相关因子mRNA表达的影响
The effects of sodium arsenite on mRNA expression level of pigment epithelium-derived factor and apoptosis-related factors in PC12 cells
摘要目的:观察不同剂量亚砷酸钠(NaAsO2)对PC12细胞(具有神经元特性的大鼠嗜铬瘤细胞)色素上皮衍生因子(PEDF)和凋亡相关因子mRNA表达的影响。方法选择PC12细胞作为研究对象,给予不同剂量的NaAsO2[0(对照组)、2、5、10μmol/L]染毒24 h,使用Real-time荧光定量PCR检测不同剂量NaAsO2染毒后PC12细胞的PEDF及其凋亡相关因子Bax和Bcl-2 mRNA的表达水平。结果 PEDF mRNA表达组间比较差异有统计学意义(F=102.28,P<0.05),其中2、5、10μmol/L NaAsO2染毒组的PEDF mRNA表达(0.70±0.07、0.33±0.04、0.23±0.10)均低于对照组(1.15±0.11,P<0.05)。 Bax mRNA表达组间比较差异无统计学意义(0、2、5、10μmol/L组分别为0.95±0.12、0.80±0.11、0.88±0.11、1.01±0.11,F=2.01,P>0.05);Bcl-2 mRNA表达组间比较差异有统计学意义(F=19.87,P<0.05),其中2、5、10μmol/L NaAsO2染毒组Bcl-2 mRNA表达(0.65±0.03、0.49±0.04、0.57±0.09)均低于对照组(0.95±0.11,P均<0.05);Bax/Bcl-2 mRNA表达比值组间比较差异有统计学意义(F=8.35,P<0.05),其中5、10μmol/L NaAsO2染毒组的Bax/Bcl-2 mRNA表达比值(1.80±0.72、1.82±0.36)高于对照组(1.02±0.24,P均<0.05)。结论 NaAsO2可能通过降低PC12细胞PEDF mRNA的表达来增加凋亡相关因子Bax/Bcl-2 mRNA表达,继而引起细胞凋亡。

The mRNA expression of PEDF and apoptosis-related factors (Bax, Bcl-2) were detected by real-time quantitative PCR.
