拓扑异构酶Ⅰ抑制剂对L1210/CDDP细胞株杀伤和诱导凋亡作用的研究
The study of killing and proapoptotic effects of topoisomerase I inhibitor on L1210/CDDP cells
摘要目的观察拓扑异构酶Ⅰ抑制剂(拓扑替康,TPT)对L1210/CDDP细胞株杀伤和诱导凋亡作用.方法采用MTT法、Annexin V染色、梯状DNA电泳与细胞形态学观察等方法分析TPT对L1210/CDDP细胞的杀伤作用、凋亡作用,分析靶细胞杀伤、凋亡作用与Caspase的关系.结果TPT对靶细胞株具有较强的杀伤作用,而且具有明显的时间依赖性,同时,靶细胞出现凋亡表现.Caspase酶特异性抑制剂对凋亡具有一定的影响.结论TPT对L1210/CDDP细胞株具有较强的杀伤和诱导凋亡作用,其机制可能涉及到Caspase-8、3的有序性活化.

Methods Analyze Topotecan' s killing activity and inductive effect on L1210/CDDP cell lines and the relationship between the above two and Caspase, using MTF method,Annexin V FITC staining, DNA ladder electrophoresis and cell morphology methods. Results TPT on L1210/CDDP cell line has strong anti-role, and a clear time-dependence. At the same time, the target cells present a sign of apoptosis. Caspase-specific inhibitor can cause a certain effect on apoptosis. Conclusion Topotecan' s killing activity and inductive effect on L1210/CDDP cell lines can result in activation of Caspase-8 and Caspase-3.
