摘要目的 探讨未感染幽门螺旋杆菌(HP)的胃癌样本与感染HP的胃癌样本之间的差异.方法 提取TCGA数据库中胃癌样本基因表达量(MRNA)的数据.根据胃癌样本中有无HP感染将样本分为2组,通过edgeR算法分别对比表达量数据,并得到差异基因.再将得到的两组差异基因分别进行富集分析,最后比较两组富集分析的结果,得出未感染HP的胃癌样本与感染HP的胃癌样本之间富集通路的差异.结果 由KEGG数据库可知,未感染HP的胃癌样本在代谢途径、蛋白质消化和吸收、脂肪消化和吸收、胃酸分泌、胰腺分泌、cAMP信号通路、血管平滑肌收缩方面与癌旁比较,差异有统计学意义(P<0.00001).感染HP的胃癌样本在细胞因子-细胞因子受体相互作用方面与癌旁比较,差异有统计学意义(P<0.00001).结论 HP感染导致细胞因子-细胞因子受体相互作用异常.可能有2种不同的胃癌亚型,分别对于代谢途径、蛋白质消化和吸收、脂肪消化和吸收、胃酸分泌、胰腺分泌、cAMP信号通路、血管平滑肌收缩方面与细胞因子-细胞因子受体相互作用方面产生影响.

abstractsObjective To investigate the difference between gastric cancer samples without helico-bacter pylori infection and gastric cancer samples infected with helicobacter pylori in gastric cancer sam-ples. Methods Data of gastric cancer gene expression( MRNA) in TCGA database were extracted. Accord-ing to the presence or absence of helicobacter pylori infection in gastric cancer samples,the samples were divided into two groups. The expression data was compared by edgeR algorithm and the differential genes were obtained. And then the two groups of different genes were analyzed by enrichment. Finally,the results of enrichment analysis were compared between the two groups to find the difference between the gastric cancer samples without helicobacter pylori infection and the gastric cancer samples infected with helicobact-er pylori infection. Results The KEGG database showed that there were significant differences in metabolic pathways,protein digestion and uptake,fat digestion and uptake,gastric acid secretion,pancreatic secretion, cAMP signaling pathway,and vascular smooth muscle contraction in gastric cancer samples without helico-bacter pylori infection(P<0. 00001). There was a significant difference in the cytokine-cytokine receptor interaction with gastric cancer samples with helicobacter pylori infection(P<0. 00001). Conclusion He-licobacter pylori infection promotes the abnormalities of cytokine-cytokine receptor interactions. There may be two different gastric cancer,respectively,for the metabolic pathway,protein digestion and absorption,fat digestion and absorption, gastric acid secretion, pancreatic secretion, cAMP signaling pathway, vascular smooth muscle contraction and cytokine-cytokine receptor Interaction has an impact.
