摘要目的 探讨Th17/白细胞介素17 (IL-17)在儿童支气管哮喘发病机制中的可能作用.方法 选取30例支气管哮喘儿童(支气管哮喘组)和20名健康儿童(对照组)为研究对象,留取外周血标本.应用流式细胞术检测支气管哮喘患儿外周血单个核细胞(PBMC)中Thl7细胞的比例,酶联免疫吸附试验(ELISA)方法检测血浆白细胞介素17 (IL-17)及免疫球蛋白E(IgE)的浓度.结果 外周血PBMC中Th17/IL-17细胞的比例在支气管哮喘组患儿[(1.83±1.01)%]高于对照组[(1.02±0.49)%],差异有统计学意义(t=3.896,P<0.01).血浆IL-17浓度在支气管哮喘组患儿为(34.23±4.88) ng/L,显著高于对照组(19.11±3.23) ng/L,差异有统计学意义(t=6.261,P<0.01).支气管哮喘组患儿血浆IgE(399.4±45.1) ng/L显著高于对照组(58.2±19.7)ng/L,差异有统计学意义(t=7.244,P<0.01).血浆IL-17的水平与PBMC表达Th17细胞的比例呈正相关(r=0.882,P<0.01),但与血浆IgE水平无相关性(r =0.375,P<0.05).结论 Th17细胞与儿童支气管哮喘的发生具有相关性,其可能是通过分泌以IL-17为主的细胞因子来上调气道炎症反应促进支气管哮喘的发生发展.

abstractsObjective To explore the role of Th17/IL-17 in the pathogenesis of asthma in children.Methods Thirty asthma children (asthma group) and 20 healthy volunteers (control group) were selected as our subjects.Percentage of Th17 cell in peripheral blood mononuclear cell (PBMC) of asthma children was detected by flow cytometry,the level of IL-17 and IgE in plasma were examined by enzyme linked immunosorbent assay.Results Proportion of Th17 cells in PBMC of children in asthma group was (1.83 ± 1.01) %,significantly higher than that in control group ((1.02 ± 0.49) % ; t =3.896 ; P < 0.01).The IL-17 level in control group was (19.11 ± 3.23) ng/L,significant lower than that in asthma group ((34.23 ± 4.88) ng/L;t =6.261 ;P < 0.01)).The Ig E level in asthma group was (399.4 ± 45.1) ng/L,significantly higher than in control group((58.2 ± 19.7) ng/L;t =7.244,P <0.01).The level of IL-17 in plasma had a postive relationship with the proportion of Th17 cells(r =0.882,P <0.01),but not relationship was seen with the level of IgE (r =0.375,P < 0.05).Conclusion Th1 7 cell is associated with asthma development of children,and the reason may due to probably aggravating asthma condition through increasing inflammation secreting IL-17.
