高压氧对大鼠脑缺血再灌注损伤后神经干细胞的影响
Effect of hyperbaric oxygen on neural stem cell proliferation following ischemia-reperfusion injury in rats
摘要目的 探讨高压氧(HBO)对成年大鼠脑缺血再灌注损伤后神经干细胞的影响.方法 将SD大鼠随机分为假手术组(SS组),缺血再灌注组(IR组),高压氧治疗组(HBO组).采用线栓法制作大鼠大脑中动脉栓塞(MCAO)模型,持续缺血1.5 h后再灌注,分别在第4、11、31天用氯化三苯四氮唑(TTC)染色,观察梗死灶面积的变化.采用免疫组化染色显示5-溴脱氧尿核苷(Brdu)阳性细胞和巢蛋白(nestin)阳性细胞,光镜下观察并统计分析HBO对脑缺血再灌注损伤后神经干细胞的影响.结果 TTC染色显示,HBO组各时间点梗死灶较IR组均有所减小;IR各组与SS组相比,皮层和纹状体BrdU阳性细胞和nestin阳性细胞数目均显著增多(P<0.05);HBO组与IR组相比,在第11天和第31天BrdU阳性细胞和nestin阳性细胞数均显著增多(P<0.05).结论 HBO对成年大鼠脑缺血再灌注损伤后神经干细胞的增殖有促进作用.

abstractsObjective To investigate effect of hyperbaric oxygen (HBO) on neural stem cell proliferation in rats following ischemia-reperfusion injury.Methods SD rats were randomly divided into 3 groups:the sham surgery (SS) group,the ischemia-reperfusion (IR) group and the HBO treatment group.Middle cerebral artery occlusion (MCAO) was performed to develop the animal model.Reperfusion was made following 1.5-hour ischemia.Then,changes in the infarction area were observed respectively on day 4.11 and 31 with tri-phenyltetrazolium chloride (TTC) staining.After immunohistechemical staining,light microscopy was used to obtain numbers of 5-bromodeoxyuridine (BrdU) and nestin positive cells in cortex and striatum areas.Results TTC staining revealed that the infarction area of the HBO group at various time points was obviously smaller than that of the IR group.When compared with those of the SS group.numbers of BrdU and nestin positive cells in cortex and striatum all increased significantly in the IR group (P<0.05).And the numbers of BrdU and nestin positive cells increased markedly on day 11 and 31,following HBO therapy,when compared with those of the IR group (P<0.05).Conclusions HBO could effectively promote proliferation of neural stem cells in rats following ischemia-reperfusion injury.
