高压氧预处理对高原肺水肿大鼠肺水通道蛋白-1的影响
Effect of hyperbaric oxygen preconditioning on the expression of lung aquaporin-1 in the rat model of high altitude pulmonary edema
摘要目的 观察高压氧(HBO)预处理对高原肺水肿(high-altitude pulmonary edema,HAPE)大鼠肺水通道蛋白-1(aquaporin-1,AQP-1)的影响.方法 38只Wistar大鼠按数字表法随机分为对照组(C组)、HAPE模型组(HAPE组)及高压氧预处理组(HBO组).制作HAPE模型后,分别以Western Blot法及实时PCR法检测各组大鼠肺AQP-1蛋白、基因表达,同时观察肺水含量及肺病理变化.结果 HAPE组肺湿/干重比明显高于C组;HBO组肺损伤积分明显低于HAPE组;HAPE组AQP-1及AQP-1mRNA表达较C组显著降低,HBO组AQP-1及AQP-1mRNA表达均明显高于HAPE组.结论 AQP-1表达降低可能是HAPE的发病机制;HBO预处理能上调AQP-1表达,具有预防和减轻HAPE的作用.

abstractsObjective To investigate the effect of hyperbaric oxygen preconditioning on the expression of lung aquaporin-1 in the rat models of high altitude pulmonary edema. Methods Thirty-eight Wistar rats were randomly divided into 3 groups: the control group (the C group ),the HBO group and the high-altitude pulmonary edema group (the HAPE group). Western blot and real-time PCR were applied to determine the expression of AQP-1 in the lung of animals in each group. The wet-to-dry weight ratio ( W/D weight ratio) and morphology of the lung were also examined. Results Experiments showed that the W/D weight ratio of the HAPE group was obviously higher than that of the C group,lung injury scores of the HBO group were much lower than those of the HAPE group,AQP-1 and AQP-1 mRNA for the HAPE group decreased markedly at the protein and gene level than those of the C group,and that AQP-1 and AQP-1 mRNA for the HBO group were significantly higher than those of the HAPE group. Conclusions Decreased expression of AQP-1 might be involved in the mechanism of HAPE incidence. HBO preconditioning could positively regulate the expression of AQP-1 and at the same time prevent and alleviate effects of HAPE. Our study might be the first one introducing HBO preconditioning in the prevention of HAPE,and the first investigation on the expression of AQP-1 in a rat model of HAPE.
