高压氧治疗对急性一氧化碳中毒大鼠行为学和脑组织抗氧化能力的影响
Effects of hyperbaric oxygen on cognitive function and antioxidant capacity of the brains in the rats with acute carbon monoxide poisoning
摘要目的 探讨高压氧对急性一氧化碳(CO)中毒大鼠行为学变化和脑组织抗氧化能力的影响.方法 66只SD大鼠按照随机数字表法分为对照组、CO组和高压氧(HBO)组.通过旷场实验和高架十字迷宫实验观察大鼠行为学变化,通过分光光度法分别测定各组大鼠脑组织总超氧化物歧化酶(T-SOD)、铜锌超氧化物歧化酶(CuZn SOD)活性和丙二醛(MDA)含量.结果 CO组大鼠在旷场实验中15 min内中央区路程[(134±87) cm]与对照组[(33±31) cm]和高压氧治疗组[(22±21) cm]相比,均显著增加,差异有统计学意义(P<0.05);中央区停留时间[(26±13)s]与对照组[(7±7)s]和HBO组[(4±3)s]相比,均显著延长,差异有统计学意义(P<0.05).CO组在高架十字迷宫中5 min内开臂滞留时间[(30.46±17.33)s]和开臂路程[(14.91±10.59) cm]与对照组[(66.07 ±27.68)s]和[(32.58 ±7.95)cm]相比明显缩短,差异有统计学意义(P<0.05),与HBO组[(76.21±32.05)s]和[(35.29 ±20.64) cm]相比明显降低,差异有统计学意义(P<0.05).CO中毒组脑组织的T-SOD和CuZn SOD均显著低于对照组和HBO组(P<0.05),CO中毒组脑组织MDA含量明显升高(P<0.05),HBO治疗后脑组织MDA含量降低(P<0.05).结论 HBO治疗可改善由CO中毒所致的行为学异常.急性CO中毒后抗氧化能力降低,而给予HBO治疗后,脑组织的抗氧化能力显著提高.

abstractsObjective To explore the effects of HBO on cognitive function and antioxidant capacity of the brains in the rats with acute carbon monoxide (CO) poisoning.Methods Sixty-six Sprague-Dawley (SD) male rats were randomly divided into 3 groups:the normal control group,the CO intoxication group and the HBO group.Open field test and elevated cross maze were used for the monitoring of the changes in behavioral activity.The levels of T-SOD,CuZn SOD activity and MDA content in the brain tissues of various groups were detected by spectrophotometry.Results As compared with those of the control group [(33 ± 31) cm] and the HBO group [(22 ±21)cm],the distance covered in the central zone within 15 minutes in the open field test by the CO group (134 ± 87) cm was significantly longer,and statistical significance could be noted,when comparisons were made between them (P < 0.05).The duration of stay in the central zone for the CO group (26 ± 13)s was significantly prolonged,when compared with those of the control group (7 ± 7)s and the HBO group (4 ± 3) s,also with statistical significance (P < 0.05).Compared with those of the control group (66.07 ±27.68) s and (32.58 ±7.95) cm and the HBO group (76.21 ±32.05) s and (35.29±20.64) cm,the open time stay (30.46 ± 17.33) s and open time distance (14.91 ± 10.59) cm were obviously shortened,also with statistical significance (P < 0.05).T-SOD activity and CuZn SOD activity in the CO group were all significantly lower than those of the control group and the HBO group (P < 0.05).MDA level for the rats of the CO group was significantly increased,while the level of HBO group was decreased (P < 0.05).Conclusions HBO therapy could improve the abnormal behavior of the rats induced by acute CO poisoning.Acute CO intoxication could decrease the antioxidant capacity of the rats,but the antioxidant capacity could obviously be improved following HBO therapy.
