超早期高压氧处理对持续脑缺血大鼠血脑屏障通透性的影响

Effect of ultra early hyperbaric oxygen treatment on the permeability of blood-brain barrier in persistent cerebral ischemic rats

摘要目的 探讨超早期高压氧(hyperbaric oxygen,HBO)处理对持续脑缺血大鼠血脑屏障通透性的影响.方法 健康雄性SD大鼠66只,按数字表法随机分为11组,即假手术组和脑缺血2、4、6、12、24h组及HBO处理2、4、6、12、24 h组,每组6只.脑缺血及HBO组大鼠制备Longa线栓法大脑中动脉栓塞模型.HBO组大鼠造模后10 min内行HBO (0.2 MPa,1 h)处理.伊文思蓝渗透试验检测各组血脑屏障损伤程度.结果 脑缺血组大鼠在缺血4h时缺血侧血脑屏障通透性达高峰(6.15±1.81),与脑缺血组(6.15±1.81,4.64±0.71)相比,HBO组大鼠在脑缺血4h和12 h时血脑屏障通透性(3.84±1.26,3.27±0.28)降低最明显,差异均有统计学意义(均P<0.05).结论 超早期HBO处理能减少持续脑缺血大鼠早期脑组织伊文思蓝渗透量,改善血脑屏障通透性,对血脑屏障有保护作用,在脑缺血4h和12 h时血脑屏障通透性降低最明显.

abstractsObjective To explore the effect of ultra early hyperbaric oxygen (HBO) treatment on the permeability of blood-brain barrier in persistent cerebral ischemic rats.Methods 66 healthy SD rats were randomly divided into 11 groups:i.e.the sham surgical group,the cerebral ischemia for 2,4,6,12,24 hours groups and the HBO treatment for 2,4,6,12,24 hours groups,each consisting of 6 rats.The rat model of middle cerebral artery occlusion (MCAO) was developed in the rats of the cerebral ischemia groups and the HBO groups by Longa suture method.Ten minutes after development of the model,the rats in the HBO groups received HBO treatment at a pressure of 0.2 MPa (2.0 ATA) for one hour.The severity of blood-brain barrier damage in the rats of the groups was measured by Evans blue extravagating experiment.Results The permeability of blood-brain barrier reached peak at hour 4 in the rats of the cerebral ischemia group (6.15 ± 1.81).As compared with that of the cerebral ischemia group(6.15 ± 1.81,4.64 ±0.71),the permeability of blood-brain barrier in the rats of the HBO group(3.84 ± 1.26,3.27 ± 0.28) decreased most significantly at hour 4 and hour 12,and there was statistical significance when comparisons were made between the groups(P <0.05).Conclusion Ultra early HBO treatment could reduce the permeability of Evans blue extravasation in the brain tissue of the rats with persistent cerebral ischemia,improve blood-brain barrier permeability and protect the blood-brain barrier.The permeability decreased most significantly in the cerebral ischemia rats at hour 4 and hour 12.
