组织芯片技术研究FSCN1蛋白在垂体腺瘤中的表达及意义
Study on the expression and significance of FSCN 1 protein in pituitary adenomas with tissue microarray
摘要目的 探讨Fascin-1(FSCN1)蛋白在垂体腺瘤组织中的表达变化,分析其与临床特征及预后的关系.方法 采用组织芯片构建包含10例正常垂体组织和212例垂体腺瘤.垂体腺瘤按照是否侵袭分为侵袭组(97例)和非侵袭组(115例).利用Leica BOND-Ⅲ系统检测FSCN 1蛋白的表达情况.分析212例垂体腺瘤患者的临床资料,并电话随访.结果 正常垂体核阳性比例为1/10;垂体腺瘤核阳性率为34.9%(74/212),胞质阳性率为20.8% (44/212).其中侵袭组FSCN1核阳性率为54.6%(53/97),非侵袭组FSCN1核阳性率为18.3%(21/115) (x2 =30.65,P=0.004);侵袭组FSCN1胞质阳性率为37.1% (36/97),非侵袭组FSCN1胞质阳性率为6.9%(8/115) (x2=29.09,P=0.000).在67例FSCN1高表达组中,肿瘤复发率为22.4% (15/67),在145例FSCN1低表达组中,肿瘤复发率为7.6% (11/145)(x2=9.3310,P=0.002).结论 FSCN1蛋白的表达水平同垂体腺瘤是否侵袭呈密切相关,是预测垂体腺瘤复发的候选基因.

abstractsObjectives To investigate the expression changes of Fascin-1 (FSCN1) protein in pituitary adenomas and to analyze its relationship between clinical features and prognosis.Methods Tissue microarray was used to construct 10 normal pituitary tissues and 212 pituitary adenomas.The pituitary adenomas were divided into invasive (n =97) and noninvasive pituitary adenomas (n =115)according to whether they were invasive or not.Leica BOND-Ⅲ system was used to detect the expression of FSCN 1 protein.The clinic data of 212 patients with pituitary adenoma were analyzed,and they were followed up by telephone.Results The nuclear positive rate of normal pituitaries was 10.0% (1/10) ; the nuclear positive rate of pituitary adenomas was 34.9% (74/212),and the cytoplasmic positive rate was 20.8% (44/212).The nuclear positive rate of FSCN1 in the invasive group was 54.6% (53/97),that in the non-invasive group was 18.3% (21/115) (x2 =30.65,P =0.004) ; the cytoplasmic positive rate of FSCN1 in the invasive group was 37.1% (36/97),and that in the noninvasive group was 6.9% (8/115) (x2 =29.09,P=0.000).In the FSCN1high expression group,the tumor recurrence rate of 67 patients was 22.4% (15/67).In the FSCN1 low expression group,the tumor recurrence rate of 145 patients was 7.6% (11/145) (x2 =9.3310,P =0.002).Conclusions The expression level of FSCN1 protein is closely associated with whether pituitary adenoma invasion or not.It is a candidate gene for predicting the pituitary adenoma recurrence.
