胆道梗阻后肝细胞线粒体钙含量改变及粉防己碱的保护作用

摘要目的::了解胆道梗阻后肝细胞线粒体钙稳态改变及粉防己碱对肝操作的保护作用。方法::结扎、切断大鼠胆总管复制胆道梗阻模型。结果::胆道梗阻后,肝细胞线粒体钙含量随梗阻时间延长而进行性升高, P<0.01;血清T-Bil、ALT、ALP、GGT水平逐渐升高, P<0.05。坏死、炎性反应及肝纤维化程度明显减轻。线粒体钙含量与血清ALT、ALP、GGT含量变化呈显著正相关, P<0.01。 结论::肝细胞线粒体钙超载可能是胆道梗阻导致肝损伤的重要机制之一,Tet可明显减轻线粒体钙超载并对胆道梗阻所致肝损伤有明显的保护作用。

abstractsIn order ot investigate the change of mitochondrial calcium content and the protective effects of tetrandrine to hepatic injury after biliary obstruction, we performed a model of complete biliary obstruction on rats by ligating the common bile duct, With section between the two ligat urcs. Tctrudrine (Tet) was administered once a day by gavagc at a dose of 30mg/Kg. At ten, twenty and thirty days after complete biliary obstruction. The calcluj content of mitochondria, the concentration of serum T-Bil、T、ALP、GGT were monitored. Hepatic morphological changes were observed under light and transmission electron microscopy. the results showed: compared with sham operateon group, mitochondrial calcium content of CBO (complete biliary obstruclion) group increased sifnificantly, P< 0.01;serumT-Bil、ALT、ALP、GGTincreased markedly, P<0.05; hepatic injury were aggravated pro gressively under light and transmission electron microscopy. Compared.with CBO group of the same obstructed time, mitochondrial calcium contentof Tet treated groups decreased sifnificantly, P<0.01;serum T-Bil、ALT、ALP、GGT also decreased significantly, P<0.01; Degeneration and necroses of hepatocytcs, inflummutory reaction and liver fibrosis cere murkcdly lightened respectively under light and transmission electron microcopy. Mitochondrial calcium content was highly positively correlated with serum ALT、ALP, rvalues were 0.924, 0.919 respectively, P<0.01. The results suggested; mitochondrial calcium overload may be the important nechanism of hepatic injury induced by biliary obstruction, Tet can lighten Mitochondrial calcium overload and protect liver effectively against the injury due to biliary obstruction.
