SCF/c-kit基因在隐睾大鼠睾丸中的表达及其意义

Change and significance of the expression of SCF and c-kit in the experimental cryptorchidism rat testis

摘要目的 观察隐睾大鼠睾丸组织中细胞凋亡及SCF/c-kit基因及其表达产物的变化,探讨隐睾致生精细胞受损的机制.方法 30只SD大鼠随机分为对照组和实验组,实验组复制单侧(左侧)腹腔隐睾模型.1个月后分别取两组左侧睾丸组织进行real-time逆转录-聚合酶链反应(RTPCR)、Western blot及免疫组织化学检测SCF/c-kit基因及其蛋白表达变化,TUNEL法检测细胞凋亡.结果 与对照组比较实验组隐睾生精细胞凋亡明显增加,对照组与实验组凋亡指数分别为5.4±1.02、19.7±3.83,差异有统计学意义(P<0.05);实验组SCF、c-kit mRNA的表达相对含量分别为0.57±0.04、0.38±0.06,差异有统计学意义(P<0.05);SCF和c-kit蛋白在对照组和实验组相对表达含量分别为1.52±0.47、0.61±0.21;0.89±0.13、0.38±0.07,差异有统计学意义(P<0.05);SCF与c-kit均有显著相关(r=0.955,P<0.01).结论 隐睾可致睾丸生精上皮细胞凋亡增加且与SCF/c-kit基因表达异常有关.

abstractsObjective To investigate the germ cell apoptosis and the change in expression levels of SCF and c-kit in the experimental cryptorchidism rat testis. Methods 30 male SD rats were divided into two groups: control group and experimental group. The control group was treated with sham-operation on the left side testis and the experimental cryptorchidism group was treated on the same side by surgery of displacing the testis inside the abdominal cavity, followed by suture of the inguinal canal. After 1 months the rats were sacrificed and the testes were harvested. The histopathological changes were observed by microscope and the expression of SCF and c-kit were detected by immunohistochemical assay, quantitative Real-time reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting analysis. Cell apoptosis was detected by TUNEL. Results The testis weight of unilateral cryptorichid rats reduced greatly and marked germ cell loss could be seen when compared to the control group and the incidence of apoptosis in control group and experimental cryptorchidism group was 5. 4 ± 1. 02,19. 7 ± 3. 83 respectively ( P < 0. 05 ). The ckit and SCF mRNA expression was 0. 38 ± 0. 06,0. 57 ± 0. 04 respectively, decrease dramatically when compared to the control group ( P < 0. 05 ). The protein expression of SCF and c-kit were detected in both control group and experimental cryptorchidism group with 1. 52 ± 0. 47,0. 61 ± 0. 21,0. 89 ± 0. 13 ,0. 38 ±0. 07 (P < 0. 05). Conclusion The decreased expression of SCF and c-kit may play a very important role in germ cell apoptosis caused by experimental cryptorchidism.
