微小RNA-125a在骨肉瘤组织的表达及其临床意义
The expression level of microRNA-125a and its clinical significance in osteosarcoma
摘要目的 检测骨肉瘤组织及其癌旁组织中微小RNA(miRNA,miR)-125a基因表达差异,分析miR-125a表达水平差异与患者临床病理特征的相关性.方法 收集郑州市骨科医院2014年10月至2015年12月20对骨肉瘤患者癌组织及其对应癌旁组织手术标本,提取20对标本总RNA,实时定量反转录聚合酶链反应(RT-qPCR)检测骨肉瘤组织及其癌旁组织标本中miR-125amRNA表达差异,分析miR-125a表达与患者临床病理特征的相关性.结果 20对配对骨肉瘤及其癌旁组织RT-qPCR检测结果显示,与癌旁组织比较,miR-125a在12例癌组织中呈低表达,占总体样本的73.33% (12/20),癌组织中miR-125a mRNA的相对表达量为(0.4098 ±0.0673)明显低于癌旁组织(0.932 7 ±0.081 6),差异有统计学意义(t=2.920,P=0.010),且两者之间的平均表达差异达2.28倍.临床资料分析结果显示,miR-125a在骨肉瘤组织中的表达差异与骨肉瘤患者病理分期(x2=23.375,P=0.000)、是否规律化疗(x2=4.444,P=0.035)及有无肺转移(x2=4.469,P=0.043)关系密切;与年龄、性别无关(x2=0.317,P=1.000).结论 miR-125a在骨肉瘤组织中的表达明显低于其相应的癌旁组织;miR-125a与骨肉瘤的Enneking外科分期、有无肺转移及是否规律化疗显著相关.

abstractsObjective To examine the expression level of microRNA (miRNA,miR)-125a in osteosarcoma tissues and non-tumor tissues,and analyze the correlation between miR-125a expression difference and clinicopathological features of patients.Methods Twenty pairs of osteosarcoma tissues and normal bone tissues in Zhengzhou Orthopedics Hospital during October 2014 to December 2015 were investigated.The total RNA was extracted,and the expression difference of miR-125a in osteosarcoma tissues and adjacent non-tumor tissues was detected by real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR).The relationship between miR-125a expression level and clinicopathological features of patients with osteosarcoma was analyzed using Log-rank test.Results The miR-125a expression was significant decreased in 73.33% (12/20) of osteosarcoma tissues (0.409 8 ± 0.067 3) as compared with matched adjacent non-tumo tissues (0.932 7 ± 0.081 6;t =2.920,P =0.010) with an average of 2.28-fold reduction in expression.The expression difference of miR-125a in osteosarcoma tissues was significantly associated with the progression (x2 =23.375,P =0.000),regular chemotherapy (x2 =4.444,P =0.035) and poor prognosis of patients (x2 =4.469,P =0.043) with osteosarcoma,but not with sex and age (x2 =0.317,P =1.000).Conclusion Decreased miR-125 a is observed in osteosarcoma tissues,which is associated with patients' aggressive pathologic features.
