E1A因子在膀胱癌组织中的表达及其对基质金属蛋白酶-2的调控作用
Expression of E1AF in bladder carcinoma and its regulation on MMP-2
摘要目的测定人膀胱移行细胞癌组织中,不同病理分期的人细胞5型腺病毒早期转录单位E1A因子(E1AF)和基质金属蛋白酶-2(MMP-2)的表达,探讨二者之间关系及其临床意义。方法采用RT-PCR和Western Blot法,检测41例人膀胱癌组织中E1AF和MMP-2的mRNA及蛋白表达,并运用计算机图像扫描分析仪测定图像的吸光度值(A值)进行半定量分析。结果 26例表浅性膀胱癌中,E1AF的阳性率为26.90%,MMP-2和E1AF的A值分别为36.09±5.82和29.08±6.26;15例浸润性膀胱癌中,E1AF的阳性率为66.67%,MMP-2和E1AF的A值分别为119.98±10.44和89.62±10.96。E1AF的阳性率、MMP-2和E1AF的A值在两组均有显著性差异;E1AF在G3级膀胱癌组织中的表达量显著高于G1级;MMP-2和E1AF的表达呈正相关(r=0.536,P<0.01)。结论 E1AF调节MMP-2的表达;二者高表达与膀胱癌浸润有关。

abstractsObjectives To measure the expression of E1AF and matrix metalloproteinase (MMP-2) on tissues of bladder transitional cell carcinoma(BTCC) and to discuss their relationship and clinical significance. Methods 41 human BTCCs were studied by RT-PCR and Western Blot to determine the expression of mRNA and protein of E1AF and MMP-2 respectively. After that, computer-assisted image scan and half-quantitative analysis were performed. Results In 26 superficial BTCCs, the positive rate of E1AF was 26.90%, and the OD value of MMP-2 and E1AF was 36.09±5.82 and 29.08±6.26 respectively. In 15 invasive BTCCs, the positive rate of E1AF was 66.67%, and the OD value of MMP-2 and E1AF was 119.98±10.44 and 89.62±10.96 respectively. Significant differences in the OD values of E1AF and MMP-2 and positive rate of E1AF were observed between the two groups. The expression of E1AF in grade Ⅲ was higher than that in grade Ⅰ. The expression of E1AF was significantly associated with that of MMP-2 in the bladder tissues (r=0.536,P<0.01). Conclusion The expression of MMP-2 is regulated by E1AF. The high expression of E1AF and MMP-2 is associated with the invasion and prognosis of BTCC.
