摘要本文采用离子交换、凝胶过滤、SDS-聚丙烯酰胺凝胶电泳或干扰素单克隆抗体亲和层析等方法,将国产人αD型基因工程干扰素纯化1,000~2,500倍至电泳纯级,回收率为18.8~52.3%。亲和层析纯化后的干扰素比活性达1.9×10 7IU/mg蛋白,分子量为20,000道尔顿,与根据人αD型干扰素基因推算的多肽分子量相一致。本方法为批量生产临床级人rIFN-αD提供了基础。

abstractsRecombinant human interferon αD type (rIFN-αD) produced in E. coli carrying pB V 867 had been purified to homogeneity with 19~52% recovery by using several successive purification steps. The purified human interferon showed a single band of MW 20,000 on SDS-PAGE. Specific activity of the purified interferon is 1.9×10 7/mg protein.
