REGULATIVE ACTIONS OF SUBSTANCE P AND SOMATOSTATIN ON THE INTERFERON-γ PRODUCTION IN MOUSE SPLEEN CELLS INDUCED BY POKEWEED MITOGEN
摘要本实验研究了P物质(Substance P, SP)和生长抑素(Somatostatin, SOM)在体外对美洲商陆(Pokeweed mitogen,PWM)诱导小鼠脾细胞产生γ干扰素(Interferon-γ,IFN-γ)的调节作用。结果显示:0.1~10μmol/L SP对5,2.5及1.25μg/ml PWM诱生的IFN-γ效价有明显增高作用,但0.001~1μmol/L SP对10μg/ml PWM诱生IFN-γ没有影响。与SP相反,0.1~10μmol/L SOM可显著降低1,0.5及0.25μg/ml PWM诱生的IFN-γ效价。用5、2.5μg/ml PWM诱生IFN-γ时,仅10μmol/L SOM有显著降低2.5μg/ml PWM诱生IFN-γ的作用,其余不同浓度SOM则没有明显降低作用。单独加SP或SOM,而不加PWM,均未测到IFN-γ。以上结果表明:SP与SOM不能直接诱生IFN-γ,在相对低浓度PWM诱生IFN-γ时,二者表现出相互拮抗的调节作用。

abstractsThe effects of substance P (SP) and somatostatin (SOM) on the interferon-γ (IFN-γ) production in mouse spleen cells induced by pokeweed mitogen (PWM) in vitro were studied. The results showed that 0.1~10μmol/L SP could significantly enhance the titres of IFN-γ induced by 5,2.5 and 1.25μg/ml PWM; 0.001~1μmol/L SP had no effect on the IFN-γ production induced by 10μg/ml PWM. In contrast to SP, 0.1~10μmol/L SOM could significantly reduce the titres of IFN-γ induced by 1,0.5 and 0.25μg/ml PWM. While stimulated with 5 or 2.5μg/ml PWM, the titres of IFN-γ induced by 2.5μg/ml PWM was significantly reduced by 10μmol/L SOM only and the other different concentrations of SOM had no significant effect on the IFN-y production. No titre of IFN-y was detected in the presence of SP or SOM without PWM. The down-regulation of 0.1 or 1μmol/L SOM in the IFN-γproduction (stimulated with 0.5μg/ml PWM) could be counteracted by 0.01~10μmol/L SP, whereas the up-regulation of 0.1 or 1μmol/L SP in the IFN-y production (stimulated with 2μg/ml PWM) could be counteracted by 0.1~10μmol/L SOM. These data indicate that SP or SOM can not directly induce IFN-γ, and there is a mutual antagonism between SP and SOM in the regulation on the IFN-y production induced by lower concentration of PWM.
