原发性肝癌及癌旁组织HBV存在状态、HBX基因产物表达和ets-2、IGF-Ⅱ,N-ras、C-myc表达的对照实验研究
A COMPARATIVE STUDY OF THE STATES OF HBV, THE PRODUCT OF HBX GENE AND THE EXPRESSIONS OF ets-2, IGF-Ⅱ, C-myc AND N-ras IN HUMAN PRIMARY HEPATOCELLULAR CARCINOMA AND TUMOR ADJACENT TISSUES
摘要本文报告了12对原发性肝细胞癌(PHC)和癌旁组织乙型肝炎病毒(HBV)存在状态。HBX基因产物-HBxAg的表达和ets-2,IGF-Ⅱ,N-ras,C-myc表达的对照研究结果。在91.5%PHC中有HBV DNA整合,并有27.2%的病人同时伴有游离HBV DNA。在75%的癌旁组织中有HBV DNA整合,其中22.2%的病人同时伴有游离HBV DNA;另有22.2%的病人仅有游离HBV DNA,未见整合,而相应的PHC中却有整合。在四种癌基因中,至少有一种以上在PHC中表达增加。IGF-Ⅱ表达为5.0和2.0kb的胚胎型转录物。HBx-Ag在肝中表达为17和/或28kd条带,并首次在人PHC血清中检测到17kd HBxAg条带。

abstractsThis paper reports the study on the states of HBV, the product of HBX gene and the expressions of ets-2, IGF-Ⅱ, C-myc and N-ras in 12 pairs of human primary hepatocellular carcinoma (PHC) and tumor-adjacent tissues. The results showed that there were integrated forms of HBV DNA in 91.5% PHC and 75% tumor-adjacent tissues. Free forms of HBV DNA were found in 4 tumor-adjacent tissues and 3 PHC. In 2 tumor-adjacent tissues only free HBV DNA was detected while in the corresponding PHC there was integrated HBV DNA, indicating that integration had taken place in the process of PHC. At least one of the 4 oncogenes studied was seen to have been over-expressed in all 12 pairs of samples. IGF-Ⅱ was expressed as 5.0 and 2.0 Kb fetal transcripts. The product of HBX gene-HBxAg was demonstrated as 17 and/or 28 Kd bands in liver samples, but only 17 Kd in serum, that had not been reported previously. The relationship and the possible roles of HBV, HBxAg and oncogenes in the carcinogenesis of human PHC are discussed in detail.
