生物钟基因在糖尿病视网膜病变发病机制中的作用机制研究进展
Research progress on the biological clock genes and diabetic retinopathy
摘要糖尿病视网膜病变(DR)是糖尿病最常见和严重的并发症之一,是成年人视力丧失的主要原因。生物钟基因能产生昼夜节律并控制其运转,若表达异常将导致多种疾病发生。越来越多的证据表明,生物钟基因在DR发病机制及发生发展中发挥重要作用。一方面,昼夜紊乱相关行为干扰了生物钟基因的昼夜振荡,其表达水平的改变易致糖代谢调控失衡,最终增加2型糖尿病及DR发病风险。另一方面,DR患者表现出昼夜节律紊乱特点,生物钟基因可能通过影响一系列视网膜病理生理过程,调控DR发生发展。因此,维持正常的昼夜节律可作为疾病预防策略,研究生物钟基因在DR中的分子机制可为更加全面地阐述DR发病机制、寻找治疗新靶点提供新思路。

abstractsDiabetic retinopathy (DR) is one of the most common and serious complication of diabetes mellitus, which is the main cause of vision loss in adults. Biological clock genes produce circadian rhythms and control its operation, while the disorder of the expression causes the occurrence and development of a series of diseases. It has been demonstrated that biological clock genes might take effects in the development and progression of DR. On the one hand, circadian rhythm disorder-related behavior disrupts the circadian oscillation of clock genes, and the change in its expression level is prone to unbalanced regulation of glucose metabolism, ultimately increasing the risk of type 2 diabetes mellitus and DR pathogenesis. On the other hand, DR patients exhibit symptoms of circadian rhythm disorders, and it has been suggested that the clock genes may control the development and progression of DR by affecting a variety of retinal pathophysiological processes. Therefore, maintaining normal circadian rhythm can be used as a disease prevention strategy, and studying the molecular mechanism of clock genes in DR can provide new ideas for more comprehensive elaboration of the pathogenesis of DR and search for new therapeutic targets.
