沙利度胺对人结肠癌HCT-8细胞周期及NF-кB、PPARγ表达的影响
Effect of thalidomide on cell cycle and the expression of PPARγ and NF - кB of HCT - 8 cell line about human colorectal cancer
目的 通过观察沙利度胺对人结肠癌HCT-8细胞周期及其对过氧化物酶增殖物激活受体γ(PPARγ)、NF-Кb蛋白表达的影响,探讨沙利度胺在结肠癌治疗上新的作用机制.方法 流式细胞术(FCM)检测细胞周期分布,免疫组化SP法检测PPAγ、NF-Кb蛋白表达.结果 流式细胞术显示细胞周期阻滞于G0/G1期,各浓度组与对照组相比差异具有统计学意义,随着沙利度胺浓度的增大,PPARγ蛋白表达显著增加,NF-Кb则明显降低.结论 沙利度胺抑制结肠癌细胞增殖的作用,可能与PPARγ蛋白表达增加、NF-Кb表达下降有关.
