摘要目的 探讨Raf激酶抑制蛋白(RKIP)和磷酸化Raf激酶抑制蛋白(P- RKIP)在胃癌组织中的表达及其临床意义.方法 选取2002年1月至2010年1月胃癌组织及癌旁组织标本各80例.纳入标准:首次手术且术前未接受化疗和放疗的患者,具有完整的临床资料和病理资料.应用免疫组织化学方法检测RKIP和P -RKIP蛋白在胃癌组织及癌旁组织中的表达,并分析RKIP和P- RKIP蛋白的表达与临床病理资料之间的关系.结果 免疫组化结果显示胃癌及癌旁组织中RKIP的阳性率分别为20.0％、80.0％,P- RKIP为26.3％、73.7％；胃癌组织RKIP和P- RKIP染色平均值分别为0.563±0.895和0.519±0.476.癌旁组织RKIP和P- RKIP染色平均值分别为1.182±0.752和0.933±0.513.胃癌组织中RKIP和P- RKIP的表达显著低于癌旁组织,差异有统计学意义(P＜ 0.05)；RKIP和P- RKIP的表达均与胃癌淋巴结转移及组织分化程度相关,尚不能认为与性别和年龄相关(P＜0.05).结论 RKIP和P- RKIP表达的减少或缺失与胃癌的发生发展、侵袭转移密切相关.

abstractsObjective To investigate the expression of Raf kinase inhibitor protein (RKIP) and phosphorylated Raf kinase inhibitor protein (P- RKIP) in gastric carcinoma and their relation with the invasion and metastasis of gastric carcinoma.Methods Immunohistochemistry was employed to detect the expression of RKIP and P - RKIP in gastric carcinoma and paratumor tissues,respectively.Statistical analysis was used to determine the relationship between their expression level and clinicopathological parameters.Results The positive rates of RKIP in gastric carcinoma and paratumor tissues were 20.0％ and 80.0％,respectively,and that of P -RKIP were 26.3％ and 73.7％.The average staining of RKIP and P -RKIP were 0.563 ±0.895 and 0.519 ±0.476 in gastric carcinoma,and were 1.182 ±0.752 and 0.933 ± 0.513 in paratumor fissues.Statistical analysis revealed that the expression level of RKIP and P - RKIP in tumor tissue were significantly lower than that in paratumor tissues ( P ＜ 0.05 ).RKIP and P - RKIP expression were dramatically lower in gastric carcinoma with lymphatic metastasis than that without lympatic metastasis.In moderately and well differentiated gastric carcinoma both RKIP and P -RKIP positive stain rate was significantly higher than that in poorly differentiated ones.There was a positive correlation between RKIP and P - RKIP expression in gastric carcinoma and in paratumor tissues.Conclusions The expression of RKIP and P - RKIP are down - regulated in gastric carcinoma tissues.And the expression level of RKIP and P - RKIP are correlated with the differentiation and lymphatic metastasis of gastric carcinoma.
