摘要Wehaveexaminedtheeffectsofinducedautologousstemcellsonbloodsugarlevelsinarabbitmodel oftype 1 diabetes.Rabbitskinfibroblastswereinducedtodedifferentiateintomultipotentstemcells,andweretransplantedintothetreatmentgroupviathepancreaticartery.Afterthefibroblastshadbeen inducedfor72h,someofthembecamemultipotentstemcells.Fourweeksafter cell transplantation,blood glucose levels of the induced stem cell treatment group were significantly lower.Theplasma insulinandplasmaC-peptidelevelsofthetreatedgroupweresignificantlyincreased(P<0.05).Thes hapeandnumberofisletswasdifferent.Inthecontrolgroup,inducedcelltreatmentgroupandnon -inducedcelltreatmentgroup.Inthecontrolgroup,islet b-cell nucleoli were obvious, and cell volumes were larger with more abundant cytoplasm.The roughendoplasmicreticulumwaswell-developedandalargenumbero fsecretorygranulescouldbe seenwithinthecytoplasm.Intheinducedcelltreatmentgroup,isletbcellswerescattered,andtheirnu cleiwereovalandslightlyirregularinshape.Thecytoplasmo fthesecellscontainedanearlynormaln umberofsecretorygranules.Inthenon-inducedcelltreatmentgroup,isletb-cellswereatrophiedand cellvolumeswerereduced.Cytoplasmicendocrinegranulesweresignificantlyreducedorabsent.In conclusion,treatmentwithinducedmultipotentstemcells canreducebloodsugarlevels,improveisletcell function,andrepair damagedpancreas in a rabbit model of type 1 diabetes.
