摘要目的 检测细胞周期分布的改变以探讨温和性高温对鼻咽癌细胞系CNE-2Z放射增敏作用的机制.方法 实验分对照组、单纯加热组、单纯照射组和联合处理组.采用流式细胞仪分析G2/M期分布比例及凋亡百分比,Western-Blotting检测CyclinB1的表达,改良的荧光染色法观察核碎片形态.结果 联合处理组G2/M期分布比例较高,CyclinB1表达较高,并产生更多的核碎片,放射诱导的凋亡百分比并不增加.结论 G2/M期分布比例增高,CyclinB1表达增加是温和性高温对鼻咽癌细胞系CNE-2Z放射增敏作用的机制.

abstractsObjective To explore the mechanism of action of mild high temperature in radiosensitization for nasopharyngeal carcinoma cell line CNE-2Z. Methods There were control group,heating alone group, radiation alone group, and heating plus radiation group in this study. The change of distribution of G2/M phase and the apoptosis percentage of the cells were analyzed by flow cytometry (FCM).Expressions of CyclinB 1 were detected by Western blot. The morphology of nuclear debris was observed with modified fluorescent staining. Results Greater percentage of distribution of G2/M phase, higher expression of cyclinB1, and more nuclear debris were found in the heating plus radiation group. No percentage of the radiation-induced apoptosis increased. Conclusions Elevated percentage of distribution of G2/M phase and higher expression of CyclinBl is the mechanism of mild high temperature in radiosensitization for nasopharyngeal carcinoma cell line CNE-2Z.
