雾化吸入三氧化二砷及艾叶油对哮喘豚鼠白三烯C4及Th1/Th2失衡的影响
Effect of inhaled arsenic trioxide and moxa leaf oil on leukotriene C4 and Th1/Th2 imbalance in asthmatic guinea pigs
摘要目的 探讨三氧化二砷(As2O3)及艾叶油雾化吸入对哮喘豚鼠白三烯C4及Th1/Th2失衡的影响及平喘的机制.方法 采用卵白蛋白(OVA)致敏激发复制哮喘豚鼠模型,60只豚鼠随机分为As2O3雾化吸入组[B1:2.0 mg/(kg·d)、B2:4.0 mg/(kg·d)]、联合用药组[C1:2.0 mg/(kg·d)+ 0.05 ml、C2:4.0mg/(kg·d)+0.1 ml];同时设生理盐水雾化吸入组(A组)及正常对照组(D组).用药7天后,比较支气管肺泡灌洗液(BALF)中白三烯C4的变化及Th 1/Th2失衡的情况.结果 (1)LTC4:B1、B2、C1、C2与A、D组比较差异均有统计学意义(P<0.01);B1与C1组比较差异有统计学意义(P< 0.01); B2与C2组比较差异有统计学意义P(<0.05);B1与B2组比较差异无统计学意义(P>0.05).(2) Th1/Th2失衡:①IFN-γ:B1、B2、C1、C2与A、D组比较差异均有统计学意义(P<0.01),用药四组之间比较差异无统计学意义(P>0.05);②IL-4:B1与B2组比较差异有统计学意义(P<0.01),B1与C1组、B2与C2组比较差异无统计学意义(P>0.05);③IFN-γ/IL-4:B1与B2组比较差异有统计学意义(P<0.05),B1与C1组、B2与C2组比较差异无统计学意义(P>0.05).结论 白三烯C4的升高及Th1/Th2的失衡是支气管哮喘的重要发病机制,As2O3平喘的机制与降低LTC4及调节Th 1/Th2的失衡有关;艾叶油平喘的机制与降低LTC4有关,较小剂量的As2O3及艾叶油联合雾化吸入可安全有效地达到平喘的作用,二者联合应用有协同作用.
