发热伴血小板减少综合征病毒感染者血清microRNA表达谱分析

Analysis of the serum microRNAs profiling associated with severe fever with thrombocytopenia syndrome virus infection

摘要目的 了解发热伴血小板减少综合征病毒(severe fever with thrombocytopenia syndrome virus,SFTSV)感染急性期患者血清microRNAs (miRNAs)的表达谱,鉴定与病毒感染或宿主免疫应答相关miRNAs.方法 采用TaqMan低密度芯片获得血清miRNA的表达谱,分析患者与正常人miR-NA的表达差异,鉴定与病毒感染或免疫相关的miRNA,最后用实时定量RT-PCR对部分miRNA进行验证.结果 TLDA芯片结果显示,与正常人血清相比,SFTS血清表达上调超过2倍的miRNA有117个,表达下调超过2倍的miRNA有30个.在表达差异较为明显的miRNA中,病毒感染或免疫相关miRNA有23个.对6个miRNA的表达进行定量RT-PCR验证,其中miR-155,miR-126,miR-15b,miR-19b与芯片结果一致.结论 通过TaqMan低密度芯片获得了SFTS病毒感染者血清miRNA的表达谱,其中部分miRNA与病毒感染或免疫相关.

abstractsObjective To understand the serum microRNAs profiling associated with severe fever with thrombocytopenia syndrome virus (SFTSV) infection,and to identify the miRNAs related to viral infection or immune responses.Methods TaqMan Low-Density Array (TLDA) was used to analyze expression levels of miRNAs in serum samples from the patients and healthy controls.Some miRNAs associated with viral infection or immune responses were examined by quantitative reverse transcriptase polymerase chain reaction (qRT-PCR).Results Compared with the healthy controls,TLDA revealed 147 miRNAs were differentially expressed (fold-change > 2) in patients,among which 117 miRNAs were up-regulated and 30 were downregulated.Twenty-three of 147 differentially expressed miRNAs were related to viral infection and immune responses.Six miRNAs were examined by qRT-PCR,and four miRNAs (miR-155,miR-126,miR-15b,miR19b) were consistent with TLDA.Conclusions Serum microRNAs profiling associated with SFTSV infection was obtained by TLDA,and some miRNAs were related to viral infection or immune responses.
