妊娠小鼠子宫蜕膜树突状细胞生物学特征的研究
The biological characteristics of the dendritic cell in uterus decidual tissue of pregnant mice
摘要目的 通过研究妊娠小鼠子宫蜕膜DC生物学特征的改变,探讨妊娠免疫抑制机制.方法 无菌分离妊娠第10天Balb/c小鼠子宫蜕膜组织的单个核细胞、rmGM-CSF、rmIL-4诱导分化为DC,未妊娠小鼠的子宫内膜组织作为对照.流式细胞仪检测DC表面标志,MTT法检测其同种异基因淋巴细胞刺激能力,ELISA法测定细胞因子的分泌水平.结果 与对照组相比,妊娠小鼠子宫蜕膜组织CD11b+树突状细胞百分率降低,CD8a+ DC百分率升高(t=3.12,t =2.54,P均<0.01),同种细胞的刺激能力明显降低,刺激细胞∶反应细胞为1∶10、1∶20、1∶50和1∶100的P值均小于0.01(t值分别为3.36、2.53、2.42和2.12).妊娠小鼠子宫蜕膜DC产生较高水平的IL-10和较低水平的IL-12(t =2.84,t=2.46,P均<0.01).结论 妊娠小鼠子宫蜕膜DC具有不同于正常小鼠子宫内膜树突状细胞的生物学特征,这可能是妊娠免疫抑制机制之一.

abstractsObjective To explore the immunological mechanism of immunosuppression pregnancy.Methods Uterus decidual mononuclear cells of 10 day pregnant Balb/c mice isolated under sterile conditions were induced to differentiate into dendritic cells with rmGM-CSF and rmIL-4.Uterus endometrum of the unpregnant mice were used as control.The surface markers of dendritic cells were analyzed with flow cytometry.The competence of stimulating allogeneic lymphocyte was detected by methyl thiazolyl tetrazolium (MTT) assay.Cytokines were determined by enzyme linked immunosorbent assay.Results Compared with control,the dendritic cells of pregnant mice expressed lower CD11b and higher CD8a (t =3.12,t =2.54,both P <0.01).Its stimulating capability for allogeneic T lymphocytes decreased significantly(as the ratio of stimulating cell and responding cell were 1∶10,1∶20,1∶50 and 1∶ 100,t is 3.36,2.53,2.42 and 2.12 respectively,and all P < 0.01).The dendritic cells of pregnant mice secreted more IL-10 and less IL-12 compared with control(t =2.84,t =2.46,both P <0.01).Conclusion The uterus decidual dendritic cells of the pregnant mice have different biological characteristics compared with normal control,and this maybe one of the mechanism for pregnancy immunosuppression.
