甲状腺相关眼病治疗新希望——IGF-Ⅰ受体拮抗剂
New hope for the treatment of thyroid associated orbitopathy-IGF-Ⅰ receptor antagonist
摘要甲状腺相关性眼病(thyroid associated orbitopathy,TAO)也称为Graves眼病,多见于女性,是一种进行性、具有潜在致盲风险的自身免疫性疾病。既往用于中重度活动性TAO的治疗药物虽然有多种,但疗效均不够确切,不能满足临床需求。随着对TAO发病机制的研究不断深入,胰岛素样生长因子Ⅰ(insulin-like growth factor-Ⅰ,IGF-Ⅰ)受体介导通路在TAO发生、发展过程中的作用得到肯定。新型靶向药物IGF-Ⅰ受体拮抗剂(如Teprotumumab)用于TAO治疗的多项临床试验取得了良好疗效,为有效治疗TAO带来希望。

abstractsThyroid associated orbitopathy (TAO), also known as Graves′ orbitopathy, is a progressive autoimmune disease with potential blindness risk that occurs more common in women. A variety of drugs have been used in the treatment of moderate and severe active TAO in the past decades, but the benefits are not satisfactory. With the in-depth research on the pathogenesis of TAO, the role of insulin-like growth factor-Ⅰ (IGF-Ⅰ) receptor-mediated pathway in the development of TAO has attracted more and more attention. A number of clinical trials using new targeted drugs IGF-Ⅰ receptor antagonists (e.g.Teprotumumab) on the treatment of TAO have achieved good outcomes, which brings new hope for effective treatment of TAO.
