摘要染色体步移技术是一种重要的分子生物学研究技术,该技术现阶段主要分为结合基因组文库的染色体步移技术和基于PCR扩增的染色体步移技术.鉴于染色体步移技术可以克隆已知序列相邻的侧翼序列,所以主要应用于克隆已知序列的侧翼启动子和调控原件、鉴定T-DNA转座子或转座子的插入位点、探知特定位点的上下游序列和染色体人工测序中的空隙填补,从而获得完整的基因组序列.随着染色体步移技术的不断改进,其在基因组学和分子生物学中得到了广泛的应用.该文就目前染色体步移技术的应用及进展加以综述.

abstractsChromosome walking technique is an important technique in molecular biology research.Chromosome walking technique by its means is divided into genomic library combined technology and PCR-based technology.As the chromosome walking technique may clone the flanking sequences adjacent to a known sequence,this technique is mainly used in cloning the promoter of known sequence and flanking regulatory element,identifying T-DNA transposon and insertion site of the transposon,discovering upstream and downstream sequence of specific genomic site,gap filling in artificial chromosome sequencing as to obtain the complete genome sequence.As chromosome walking technology continues to improve,it is widely used in genomics and molecular biology.In this review,we demonstrate some key applications of chromosome walking techniques in recent years.
