大肠癌组织中PTEN和基质金属蛋白酶9及血管内皮生长因子C的表达及相关性研究
Correlation and expression of PTEN, matrix metalloproteinase-9 and vascular endothelial growth factor-C in colorectal cancer
摘要目的:探讨抑癌基因PTEN和基质金属蛋白酶9(MMP-9)及血管内皮生长因子C(VEGF-C)在大肠癌组织中的表达及相关性。方法应用免疫组化SP法检测102例大肠癌组织及30例正常大肠黏膜组织中PTEN、MMP-9和VEGF-C的表达,并分析其与大肠癌临床病理特征的关系。结果本组大肠癌与癌旁组织中PTEN阳性表达率分别为54.9%(56/102)和100%(30/30),MMP-9阳性表达率分别为83.3%(85/102)和21.7%(8/30),VEGF-C阳性表达率分别为69.6%(71/102)和16.7%(5/30),差异均有统计学意义(均P<0.01)。大肠癌组织中PTEN与MMP-9和VEGF-C表达呈负相关,MMP-9与VEGF-C的表达呈正相关(均P<0.05)。3者的表达均与年龄、性别、肿瘤大小等无关,而与肿瘤淋巴结转移、远处转移、分化程度及TNM分期有关(均P<0.05)。 PTEN阳性表达组术后1、3、5年生存率明显高于阴性组(均P<0.05)。 MMP-9与VEGF-C阳性表达组术后1、3、5年生存率均明显低于MMP-9与VEGF-C阴性组(均P<0.05)。结论 PTEN表达降低与MMP-9及VEGF-C表达升高可能与大肠癌的发生、发展密切相关,可作为判定大肠癌生物学行为和预后的临床参考指标。

abstractsObjective To investigate the correlation and expression of PTEN, matrix metalloproteinase-9 (MMP-9),and vascular endothelial growth factor-C(VEGF-C)in colorectal cancer.Methods The expression of PTEN, MMP-9 and VEGF-C in 102 cases of colorectal cancer tissues and 30 cases of normal colorectal tissues was detected using immunohistochemical SP method.Their correlations with cliniopathologic features were analyzed. Results The positive expression of PTEN in colorectal cancer was significantly lower than that in the controls (54.9%vs 100%, P<0.01).Both MMP-9 and VEGF-C were over expressed compared with the controls(83.3%vs 21.7%and 69.6%vs 16.7%respectively, P<0.01).The expression of PTEN was negatively correlated with that of MMP-9 and VEGF-C, and the expression of MMP-9 was positively correlated with that of VEGF-C ( P<0.05).The expressions of the three proteins in colorectal cancer tissues were not associated with age, gender, or tumor size.There were significant associations between the expression of these proteins and lymph node metastasis, distant metastasis, differentiation, and TNM staging(P<0.05).The 1, 3 and 5-year survival rates of the patients with positive PTEN were significantly higher than those of the patients with negative PTEN(P<0.05).The 1, 3 and 5-year survival rates of the patients with positive MMP-9 and VEGF-C were significantly lower than those of the patients with negative MMP-9 and VEGF-C( P<0.05) .Conclusions The decrease in the expression of PTEN and increase in MMP-9 and VEGF-C are closely correlated to the development of colorectal cancer.Their expressions may be favorable markers and useful prognostic indicators in the patients with colorectal cancer.
