白细胞介素-4和白细胞介素-13基因多态性与过敏性紫癜的相关性分析
Association of gene polymorphism in interleukin -4 and interleukin -13 gene with Henoch - Schonlein purpu-ra
摘要目的:探讨我国中部地区汉族儿童中白细胞介素(IL)-4(rs2243250)及 IL-13(rs1800925)基因多态性与过敏性紫癜(HSP)及紫癜性肾炎(HSPN)的相关性,并比较 HSP 和 HSPN 血清中2种细胞因子的差异。方法采取病例对照的方法,分别收集383例患儿作为试验组,其中 HSP 患儿320例(HSP 组),HSPN 患儿63例(HSPN 组),409例儿童为健康对照组;利用限制性片段长度多态性聚合酶链反应( PCR-RFLP)的方法测定2个基因多态性的分布;酶联免疫吸附实验(ELISA)测定血清中 IL-4、IL-13的水平,并利用 SPSS 16.0统计软件进行数据分析。结果1. IL-4基因的单核苷酸多态性(SNP)位点的多态性在健康对照组、HSP 及 HSPN 组间分布差异无统计学意义(P ﹥0.05);IL-13基因的 SNP 位点的多态性与 HSP 相关(P =0.037),且 T 等位基因频率明显高于健康对照组(P =0.027);试验组(HSP 组+ HSPN 组)中 TT 基因型比例与健康对照组比较差异无统计学意义(P =0.132);HSPN 组与健康对照组比较差异无统计学意义(P =0.487),且 HSP 组与 HSPN 组比较差异亦无统计学意义(P =0.129)。2. IL-13基因 SNP 位点多态性与血清中 IL-13水平相关,TT 基因型的患儿血清中水平高于其他基因型的患儿(P ﹤0.05);IL-4基因的3种基因型之间血清中 IL-4的水平差异无统计学意义(P ﹥0.05)。3. HSPN 组 IL-13的水平高于 HSP 组(P ﹤0.05),而血清中 IL-4的水平在2组间差异无统计学意义。结论 IL-13基因的 SNP 位点(rs1800925)的多态性与 HSP 相关,且该 SNP 位点多态性影响血清中 IL-13的水平;HSPN 患儿血清 IL-13的水平高于 HSP 组,IL-13升高可能是 HSP 发展为 HSPN 的危险因素。

abstractsObjective To investigate the association between 2 single nucleotide polymorphisms( SNPs) [rs2243250 in interleukin(IL)- 4 gene and rs1800925 in IL - 13 gene]in Henoch - Schonlein purpura(HSP)and Henoch - Schonlein purpura nephritis(HSPN)in Han Chinese children. The level of IL - 4 and IL - 13 in serum were compared between HSP and HSPN. Methods A case - control study was adopted in this research,and 383 children with HSP or HSPN were recruited in this study,409 normal children served as controls. The genotypes of 2 SNPs were detected by using polymerase chain reaction - restriction fragment length polymorphism(PCR - RFLP),and the serum levels of IL - 4 and IL - 13 in HSP and HSPN patients were detected by using enzyme linked immunosorbent assay (ELISA). All the data were analyzed by SPSS 16. 0 software. Results (1)There was no association(polymorphism of SNP rs2243250 in IL - 4 gene)in HSP group and HSPN group compared with the healthy control group(P ﹥ 0. 05);and the polymorphism of another SNP(rs1800925 in IL - 13 gene)was associated with HSP(P = 0. 037),and the fre-quency of T allele of the study group was higher than that of the healthy control group(P = 0. 027). But there was no difference between the study group(HSP group and HSPN group)and the healthy control group in frequency of TT genotype(P = 0. 132),and there was no association between HSPN group and healthy control group(P = 0. 487);also there was no association between polymorphism of this SNP in HSP group and HSPN group(P = 0. 129).(2)There was an association between polymorphism of IL - 13 gene and IL - 13 level in serum,patients with TT genotype had a higher serum IL - 13 level than any other genotypes(P ﹤ 0. 05),but there was no statistically significant difference in IL - 4 level between TT genotype and other genotypes(P ﹥ 0. 05).(3)There was an association between polymorphism of IL - 13 gene and IL - 13 level in serum patients with HSPN who had a higher level of serum IL - 13 than the patients with HSP( P ﹤ 0. 05),but there was no difference in serum IL - 4 level between HSP group and HSPN group. Conclusions The polymorphism of SNP(rs1800925 in IL - 13 gene)was associated with HSP group,and the poly-morphism of this SNP might affect the level of IL - 13 in serum. The patients with HSPN have a higher level of serum IL -13 than the patients with HSP,and high level of IL -13 may be a risk factor in the progression from HSP to HSPN.
