换一批高效液相色谱法同时测定竹根七中5种甾体皂苷的含量
Simultaneous determination of 5 steroidal saponin components in Rohdea chinensis Baker.by high-performance liquid chromatography
摘要目的 建立高效液相色谱法(high-performance liquid chromatography,HPLC),同时测定竹根七中5种甾体皂苷成分的含量.方法 采用 Thermo HyPURITY C18 色谱柱(250 mm×4.6 mm,5 μm);以乙腈-水为流动相,梯度洗脱;流速为1.0 mL·min-1;检测波长为 203 nm;柱温为 25℃;进样量为 10 μL.结果 竹根七中 5种甾体皂苷tupisteroide B、(25R)-26-O-β-D-glucopyranosyl-furost-1β,3β,26-trihydroxy-5(6),20(22)-dien-3-O-β-D-glucopyranoside、spirost 25(27)-ene-1β,2β,3β,4β,5β,6β,7α-heptol、aspidoside A、1β,2β,3β,4β,5β,7α-hexahydroxy-spirost-25(27)-ene-6-one的线性范围分别为 0.06～1.50、0.06～1.08、0.58～3.88、0.07～2.25、1.54～45.24 μg(r≥0.999 6);平均加样回收率依次为98.59％、99.25％、99.44％、98.33％、99.63％,相对标准偏差(relative standard deviation,RSD)分别为1.07％、0.86％、1.42％、0.90％、1.74％.结论 建立的含量测定方法简便可靠、重复性良好,可为竹根七的质量控制及开发利用提供科学依据.
