mTOR、p70S6K在体外培养血管瘤血管内皮细胞中的表达及意义
The expression of mTOR and p70S6K in hemangioma vascular endothelial cell in vitro
摘要目的 观察mTOR、p70S6K在体外培养血管瘤血管内皮细胞中的表达,探讨其与血管瘤血管内皮细胞周期分布的关系.方法 用组织块法培养血管瘤内皮细胞,同步化培养血管瘤内皮细胞后,采用Western blot法检测血管瘤内皮细胞mTOR、p70S6K-α的表达水平,同期用流式细胞仪检测血管瘤血管内皮细胞周期的变化;并分析mTOR、p70S6K-α的表达水平与血管瘤血管内皮细胞周期分布的关系.结果 mTOR蛋白表达水平较高0.20±0.01,p70S6K-α蛋白表达水平较低时0.25±0.01,处于G0/G1期的细胞较少(55.95±4.38)%; mTOR蛋白表达水平下降至0.16±0.02,p70S6K-α蛋白表达水平升高至0.29±0.01,处于G0/G1期的细胞明显增多(77.86±8.18)%;二者比较差异有统计学意义(t=- 5.781,P<0.01).结论 mTOR和p70S6K参与了体外培养的血管瘤血管内皮细胞细胞周期G0/G1阻滞.

abstractsObjective To study the effect of mammalian target of rapamycin(mTOR) and p70 ribosomal S6-kinase(p70S6K)for cell cycle in hemangioma vascular endothelial cell in vitro.Methods The hemangioma vascular endothelial cells which came from the sterile hemangioma were cultured by tissue culture method.The expression of the factor Ⅷ related antigen which only exists in the endothelial cells to identify the hemangioma vascular endothelial cell (VEC) was observed by the immunohistochemistry technique Synchronized cultured endothelial cells,mTOR and p70S6K-α.of them were detected by Western blot,and cell cycle was examined by flow cytometry technique Results The percent of vascular endothelial cell was (55.95 ± 4.38)% in the G0/G1 phase with lower mTOR expression and higher p70S6k-α expression,but it increased (77.86 ± 8.18)% with higher mTOR expression and lower p70S6k-α expression.The percent of vascular endothelial cell was sigificantly different between two groups (t =- 5.781,P<0.01).Conclusions mTOR/p70S6K may play an important role in G0/G1 phase arrest of hemangioma vascular endothelial cell in vitro.
