Application of the Bicinchoninic Acid Method to Quantitatively Detect Residual Medical Multi-Enzyme Detergent
摘要目的::建立医用多酶清洗剂的定量检测方法。方法::实验研究。应用二喹啉甲酸(BCA)法建立医用多酶清洗剂的定量检测方法,并测定其重现性、准确性、回收率。结果::建立了BCA法测定医用多酶清洗剂的标准曲线、方程及样品的数据处理方法。BCA标准曲线的线性关系良好,且多次实验稳定性佳, R2均>0.95;标准蛋白回收试验,回收率<100±15%,标准偏差(RSD)<3%;BCA法测定医用多酶清洗剂原液及1∶50、1∶100比例稀释的医用多酶清洗剂样品,RSD均<5%;将医用多酶清洗剂原液稀释成6个浓度梯度,BCA法测得在1∶50~1∶200稀释范围内,随着稀释比例增加,酶蛋白浓度呈线性下降趋势。 结论::BCA法可用于医用多酶清洗剂的定量检测,且具有良好的灵敏度、回收率、稳定性和精确性。

abstractsObjective::To establish a quantitative determination method for medical multi-enzyme detergent.Methods::This was an experimental study. A quantitative method for the determination of medical multi-enzyme detergent was established by using the bicinchoninic acid (BCA) method, and its reproducibility, accuracy and recovery were determined.Results::The standard curve, equation and the data analysis method of the samples were established. The BCA standard curve was very good and has been repeated in experiments, R2>0.95. The standard protein recovery test showed that the recovery rate was <100±15% and the relative standard deviation (RSD) was <3%. BCA method was used to determine the multi-enzyme detergent and multi-enzyme detergent of the 1∶50 and 1∶100 dilutions, RSD<5%. The multi-enzyme detergent stock solution was diluted into six concentration gradients. The results of BCA showed that the concentration of enzyme protein decreased with an increase in the dilution ratio in the range of 1∶50-1∶200. Conclusions::The BCA method can be used for the quantitative determination of multi-enzyme detergents for medical devices with good sensitivity, recovery, stability and accuracy.
