小神经胶质细胞对湿性年龄相关性黄斑变性患者脉络膜新生血管膜形成的作用
The role of microglial activation in choroidal neovascular membranes of age-related macular degeneration patients
摘要目的 探讨小神经胶质细胞在湿性年龄相关性黄斑变性(AMD)患者脉络膜新生血管膜(choroidal neovascular membranes,CNM)形成中的作用.方法 选取11例湿性AMD,利用其手术切除的CNM,石蜡包埋后切片,分别用抗HLA-DR抗原、CD 45、CD 68的抗体对小神经胶质细胞进行免疫染色.同时用抗CD 68抗体和抗血管内皮生长因子(VEGF)抗体对CNM进行双重免疫染色.结果 在炎症活跃的CNM中,可见大量阿米巴样小神经胶质细胞,胞体肥大,突起短小,并可见新鲜出血.在炎症非活跃的CNM中,可见分叉较多的小神经胶质细胞,胞体小、突起呈细长的结节状且数量多.双染的结果显示VEGF主要表达在小神经胶质细胞.结论 小神经胶质细胞存在AMD病程的各个阶段,小神经胶质细胞活化是该病的重要病理特征.

abstractsObjective To investigate the role of microglial activation in choroidal neovascular membranes(CNM) of age-related macular degeneration (AMD) patients.Methods Paraffin-embedded sections of CNM excised from 11 patients with wet AMD were collected.The microglial of sections were analyzed by immunohistochemistry with antibodies against HLA-DR antigen,CD 45 or CD 68.CNM were also dual labeled with CD 68 and vascular endothelium growth factor (VEGF).Results In inflammatory active CNM,abundant microglial cells were appeared characteristically amoeboid,with hypertrophic cell body and shorter process.In inflammatory inactive CNM,most of positive labeled microglial cells showed ramified,with small cell body and numerous nodular and slender processes.Double labeling study demonstrated that VEGF was mainly expressed in microglial cells.Conclusion Microglial are involved at different stages of the disease.Microglial activation is a prominent feature of the disease process.
