Immunomodulatory mechanisms of JAK/STAT signaling pathways in bone development and remodeling:recent advances
摘要The Janus kinase(JAK)-signal transducer and activator of transcription(STAT)signaling pathway is an important downstream mediator of multiple cytokines,hormones,and growth factors.The downstream mediated by JAK/STAT involves processes such as hematopoiesis,tissue repair,immune adaptation,inflammation,apoptosis,and adipogenesis.The absence or mutation of JAK/STAT components is associated with many diseases.Studies have shown that the JAK/STAT signaling pathway plays an important role in bone development,bone metabolism,and fracture healing.Among them,multiple inflammatory cytokines are involved in this process,mainly affecting normal bone remodeling by influencing the differentiation and maturation of osteoclasts and osteoblasts at intra-articular and extra-articular skeletal sites.This article systematically elaborates the role of the JAK/STAT pathway in skeletal development,homeostasis,and remodeling by summarizing the skeletal phenotypes of JAK/STAT knockout mouse models,the selective inhibition of JAK/STAT signal components,and the effects of JAK inhibitors on osteoclasts,osteoblasts,and osteocytes in inflammatory and non-inflammatory states.
