病理性瘢痕患者HIF-1α、VEGF、MMP-2的表达及临床意义
Expression and clinical significance of HIF-1, VEGF and MMP-2 in patients with pathological scar
摘要目的 探讨缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)、基质金属蛋白酶2(MMP-2)在病理性瘢痕的表达水平及意义.方法 选取2015年3月至2017年2月本院收治的病理性瘢痕患者73例,其中增生性瘢痕组42例,瘢痕疙瘩组31例.另选取非病理性瘢痕患者20例和正常整形者20例作为对照.采用实时荧光定量及蛋白免疫印迹技术检测各组皮肤组织中HIF-1α、VEGF、MMP-2的mRNA及蛋白表达水平,并分析其临床意义.结果 与非病理性瘢痕组织和正常皮肤组织比较,病理性瘢痕组织中HIF-1α、VEGF、MMP-2的mRNA及蛋白表达水平均明显升高,差异均有统计学意义(P<0.05).瘢痕疙瘩组中HIF-1 αmRNA和蛋白的表达水平明显高于增生性瘢痕组(P<0.05);瘢痕疙瘩组和增生性瘢痕组中VEGF mRNA和蛋白的表达水平比较差异无统计学意义(P>0.05);瘢痕疙瘩组中MMP-2 mRNA和蛋白的表达水平低于增生性瘢痕组(P<0.05).Pear-son相关分析显示病理性瘢痕组织中HIF-1 α与VEGF、MMP-2均呈正相关(r=0.623、0.507,P<0.05).结论 HIF-1α、VEGF、MMP-2在病理性瘢痕组织中的表达明显升高,与病理性瘢痕的形成密切相关.

abstractsObjective To investigate the expression levels and significance of hypoxia inducible factor-1 α (HIF-1 α),vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2) in pathological scar.Methods 73 patients with pathological scar from March 2015 to February 2017 in our hospital were selected,including 42 cases in hypertrophic scar group and 31 cases in keloid group.20 cases of non pathological scar patients and 20 cases of normal plastic surgery were selected as control group.mRNA and protein expression levels of HIF-1,VEGF and MMP-2 in skin tissue of each group were detected by real-time fluorescence quantitative and Western blot,and the clinical significance was analyzed.Results Compared with non pathological scar tissue and normal skin tissue,the mRNA and protein expression levels of HIF-1α,VEGF and MMP-2 in pathological scar tissue were significantly increased,and the differences were statistically significant (P < 0.05).The mRNA and protein expression levels of HIF-1α in keloid group were significantly higher than those in hypertrophic scar group (P < 0.05);there was no significant difference in the expressions of mRNA and protein of VEGF between keloid group and hypertrophic scar group (P > 0.05);the mRNA and protein expression levels of MMP-2 in keloid group were significantly lower than those in hypertrophic scar group (P < 0.05).Pearson correlation analysis showed that there was a significant positive correlation between HIF-1α and VEGF,MMP-2 in pathological scar tissues (r =0.623,0.507,P < 0.05).Conclusions The expressions of HIF-1α,VEGF and MMP-2 in pathological scar tissue is obviously increased,which is closely related to the formation of pathological scar.
