下咽癌组织长链非编码RNA HOXA11-AS、LEF1-AS1表达与预后的关系
Relationship between the expressions of long noncoding RNA HOXA11-AS and LEF1-AS1 in hypopharyngeal carcinoma tissues and prognosis
摘要目的:探讨长链非编码RNA(lncRNA)同源异形盒基因A11反义RNA(HOXA11-AS)、淋巴增强因子1反义RNA1(LEF1-AS1)在下咽癌组织中的表达及与预后的关系。方法:前瞻性选取2019年2月—2021年2月期间在邯郸市中心医院接受治疗的80例下咽癌患者,取其手术切除的下咽癌组织与癌旁正常组织(距癌组织边缘>2 cm)。采用实时荧光定量聚合酶链式反应(RT-qPCR)检测HOXA11-AS、LEF1-AS1的表达。比较下咽癌组织与癌旁正常组织HOXA 11-AS、LEF1-AS1的表达;分析其与临床病理特征的关系;采用Kaplan-Meier法分析HOXA11-AS、LEF1-AS1的高/低表达与下咽癌患者预后的关系。结果:下咽癌组织HOXA11-AS、LEF1-AS1表达均高于癌旁正常组织(均 P<0.05);下咽癌组织HOXA11-AS、LEF1-AS1表达与TNM分期、分化程度、淋巴结转移有关(均 P<0.05);下咽癌组织HOXA11-AS、LEF1-AS1高表达者3年总体生存率均低于低表达者(均 P<0.05)。 结论:HOXA11-AS、LEF1-AS1在下咽癌组织中表达升高,且与下咽癌患者预后不良有关。

abstractsObjective:To explore the expressions of long noncoding RNA (lncRNA) homeobox A11 antisense RNA (HOXA11-AS) and lymphoid enhancer-binding factor 1 antisense RNA 1 (LEF1-AS1) in hypopharyngeal carcinoma tissues and their relationships with prognosis.Methods:Prospectively, 80 patients with hypopharyngeal carcinoma who were treated in Handan Central Hospital from February 2019 to February 2021 were selected. The hypopharyngeal carcinoma tissues resected surgically and the adjacent normal tissues (more than 2 cm away from the edge of the cancer tissue) were obtained. The expressions of HOXA11-AS and LEF1-AS1 were detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR). The expressions of HOXA11-AS and LEF1-AS1 in hypopharyngeal carcinoma tissues and adjacent normal tissues were compared. The relationships between their expressions and clinicopathological features were analyzed. The Kaplan-Meier method was used to analyze the relationships between high/low expressions of HOXA11-AS and LEF1-AS1 and the prognosis of patients with hypopharyngeal carcinoma.Results:The expressions of HOXA11-AS and LEF1-AS1 in hypopharyngeal carcinoma tissues were higher than those in adjacent normal tissues (all P<0.05). The expressions of HOXA11-AS and LEF1-AS1 in hypopharyngeal carcinoma tissues were related to tumor node metastasis (TNM) stage, degree of differentiation, and lymph node metastasis (all P<0.05). The 3-year overall survival rates of patients with high expressions of HOXA11-AS and LEF1-AS1 in hypopharyngeal carcinoma tissues were lower than those of patients with low expressions (all P<0.05). Conclusions:The expressions of HOXA11-AS and LEF1-AS1 are increased in hypopharyngeal carcinoma tissues, which are related to poor prognosis of patients with hypopharyngeal carcinoma.
