周围神经损伤后急性期NGF、BDNF基因表达变化的实验研究
Experimental study of NGF and BDNF Gene expression in acute peripheral nerve injury
摘要目的 观察大鼠坐骨神经损伤急性期神经生长因子(NGF)、脑源性神经生长因子(BDNF)mRNA含量的变化.方法 取SD大鼠48只,随机均分为8组.其中7组切断右侧坐骨神经并原位缝合,另1组为健康对照组.分别于术后1,2,3,4,5,6,7 d以吻合口为中心,取上下各5 mm共1cm坐骨神经提取总RNA,采取反转录-多聚酶链反应(RT-PCR)技术检测组织中NGF、BDNF mRNA含量变化.结果 大鼠周围神经中NGF mRNA含量在正常组织有低水平表达.在损伤后1 d即明显升高,在损伤后5 d降低,随即恢复较高水平.BDNF在正常组织亦低水平表达,在损伤后4 d明显增高,5,6 d低至正常组织水平,7 d恢复较高水平.结论 大鼠坐骨神经损伤急性期NGF、BDNF基因表达变化不同步,提示不同因素在损伤后急性期NGF、BDNF含量变化中起作用.

abstractsObjective To investigate changes in gene expression of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) after acute peripheral nerve injury in rats.Methods Forty-eight SD rats were randomly divided into 8 groups.Right sciatic nerve injury model was established by transection and direct repair in rats of 7 groups.Another group of unoperated rats served as normal control.One cm of sciatic nerve segment centered at the repair site was harvested at postoperative day 1,2,3,4,5,6 and 7 respectively.Total RNA was extracted and expression of NGF mRNA and BDNF mRNA was quantified using reverse transcriptase-polymerase chain reaction (RT-PCR).Results NGF mRNA and BDNF mRNA could be detected in normal rat sciatic nerves at a relatively low level.Obvious increase of NGF mRNA expression was seen first day after sciatic nerve injury,which maintained at a higher level there after.BDNF mRNA rose to a higher than normal level 4 days after nerve injury.At postoperative day 5 and day 6 it dropped to nomal.At postoperative day 7 BDNF mRNA increased again.Conclusion Changes of NGF and BDNF expression are not synchronized after rat sciatic nerve injury.This indicated that different factors are involved in regulating NGF and BDNF mRNA content in acute peripheral nerve injury.
