纳米化姜黄素抑制大鼠跟腱粘连的实验研究
Prevention of Achilles tendon adhesion with nano-curcumin: an experimental study in rats
摘要目的 研究纳米化姜黄素对大鼠跟腱损伤后粘连的抑制作用,并初步探讨其相关机制.方法 取72只成年雄性SD大鼠,随机分为3组:健康对照组(A组),单纯姜黄素实验组(B组),纳米化姜黄素+激光照射组(C组).在肌腱缝合处3组分别应用等剂量的生理盐水、姜黄素及纳米化姜黄素,并在肌腱愈合过程的不同时间点(1、2、4、8周)对大鼠跟腱的大体形态、组织形态学、生物力学以及免疫组化等方面进行检测,并进行统计学分析.结果 C组肌腱粘连程度明显较A、B组轻;术后4周起,C组从早期炎性细胞浸润到中后期胶原纤维排列均明显优于A、B组;生物力学检测C组的肌腱滑动距离较其他两组明显增加,与A组比较差异有统计学意义(P<0.05).结论 通过以上实验结果可以证实,纳米化姜黄素+激光照射能明显地抑制肌腱早期局部炎性反应,具有显著预防肌腱粘连的作用.

abstractsObjective To study the effect of nano-curcumin on inhibiting tendon adhesion in a rat Achilles tendon injury model and to investigate the related mechanism.Methods The Achilles tendon was cut and repaired in 72 rats that were randomly divided into 3 groups:the control group (group A),the curcumin group (group B),and the nano-curcumin + laser group (group C).The same dose of normal saline,curcumin and nano-curcumin were applied to the tendon suture site,respectively.At different time points of tendon healing (1,2,4 and 8 weeks),the Achilles tendons were harvested and subject to gross morphological observation,histological,biomechanical,and immunohistochemical examinations.Statistical analysis was conducted to compare results from various groups.Results The adhesion score of tendons in the nano-curcumin + laser group in gross morphology was significantly lower than that of the control group (P < 0.05).Histologically,the inflammatory response score of tendons in the nano-curcumin group was significantly lower than that in the other two groups.Biomechanical tests revealed significantly enhanced tendon gliding distance in the nano-curcumin group comparing to that in the control group (P < 0.05).Conclusion Application of nano-curcumin and laser at the tendon repair site can significantly inhibit the early local inflammatory reactions,and prevent tendon adhesion.
