采用改良清晰脂质交换丙烯酰胺杂交精细成像相容性组织水凝胶法透明正中神经的实验研究
Visualizing median nerve fibers through modified CLARITY method: an experimental study in rat
摘要目的 采用改良清晰脂质交换丙烯酰胺杂交精细成像相容性组织水凝胶(Clear Lipidexchanged Acrylamide-hybridized Rigid Imaging-compatible Tissue-hYdrogel,CLARITY)法将正中神经透明化,观察神经纤维的分布和走行.方法 取成年雌性SD大鼠的正中神经,进行多聚甲醛固定,聚丙烯酰胺水凝胶包埋,免疫荧光标记,透明化处理,荧光显微镜观察.结果 多聚甲醛固定和水凝胶包埋后的神经组织呈乳白色,柔软富有弹性.透明化处理后的正中神经完全透明,荧光显微镜视下可清晰观察到深部纵向走行的神经纤维.结论 改良CLARITY法能有效透明化周围神经,为神经功能束分布的研究提供了新的方法.

abstractsObjective To observe the distribution and course of nerve fibers after median nerve vitrification using modified CLARITY method.Methods The median nerve of adult female SD rats was harvested,fixed with paraformaldehyde,embedded by polyacrylamide hydrogel,labeled by immunofluorescence for clearing treatment and observed by fluorescence microscopy.Results The nerve tissue fixed with paraformaldehyde and embedded by hydrogel was milky white,soft and elastic.The median nerve after tissue clearing treatment was completely transparent.The deep longitudinal running nerve fibers could be clearly observed by fluorescence microscopy.Conclusion The modified CLARITY method can effectively transparent peripheral nerve,which provides a new method for the study of distribution and function of nerve tracts.
