摘要目的 观察地塞米松对兔椎间盘髓核细胞增殖的影响,以及对髓核细胞表达增殖细胞核抗原(PCNA)的影响.方法 无菌条件下取兔椎间盘,常规分离消化髓核细胞进行培养;传代培养第2代髓核细胞7 d,随机分为4组,实验组给予l～100 nmol/L地塞米松进行培养,对照组给与等量磷酸盐缓冲液(PBS),培养5 d后收集细胞.采用逆转录-聚合酶链反应(RT-PCR)法检测地塞米松对兔髓核细胞内PCNA mRNA表达的影响;并提取蛋白用Western blot方法检测PCNA蛋白表达.结果 RT-PCR及Western blot检测结果表明,1～100 nmol/L地塞米松可增加兔髓核细胞内PCNA的表达,当地塞米松浓度为100 nmol/L时效果最显著,分别增加了0.42和1.37倍,差异有统计学意义(P<0.05).结论 地塞米松可以促进兔椎间盘髓核细胞的增殖,并可使兔髓核细胞内PCNA的表达增高.

abstractsObjective To study the proliferative effects of Dexamethasone on rabbit nucleus pulposus cell, and the expression of proliferation cell nuclear antigen ( PCNA ) of nucleus pulposus cells.Methods The intervertebral disk was extracted by sterile process, after digestion of nucleus pulposus the cells were cultured in DMEM medium. Then 1-100 nmol/L of Dexamethasone was added into cultured nucleus pulposus cells. The expression of PCNA mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) ;the expression of PCNA protein was detected by Western blotting. Results RT-PCR and Western blot results showed that 1-100 nmol/L of Dexamethasone has up-regulated the expression of PCNA,it has 0. 42 folder and 1.37 folder than control group. Conclusion Dexamethasone could promote nucleus pulposus cell proliferation and up-regulate the expression of PCNA.
